Invasion of Lymphosarcoma Cells Into the Perfused Mouse Liver

Abstract: Liver of (C57BL X DBA)F1 mice were perfused in situ with a synthetic hemoglobin-free medium, to which murine lymphosarcoma cells were added. All cells were arrested. They were only found in the sinusoids, predominantly in periportal areas. Many lymphosarcoma cells penetrated the walls of the sinusoids with protrusions that extended into and through the endothelial cells. The protrusions often also invaded hepatocytes, and some cells migrated out of the sinusoids. The percentage of the cells that penetrated end… Show more

“…This is an in situ confirmation of previous in vitro findings concerning tumor cell binding prior to cytoxocity and phagocytosis. 14 -16 The presence of a single large DiO-filled phagosome in KCs indicates whole tumor cell phagocytosis and degradation in vivo, in concordance with the literature, 7,15,35,52 and parallel to the lysosomal localization of fluorescein in KCs 24 hr after injection of fluorescein-labeled colon cancer cells. 53 Lysosomal degradation of the ingested tumor cells was demonstrated by the subsequent appearance of smaller DiO-filled vacuoles resembling lysosomes at the electron microscopy level and the complete disappearance of the phagosome after 24 hr.…”

“…The liver contains a large number of resident macrophages, the Kupffer cells. In our studies on lymphosarcoma cells (Dingemans, 1973;Roos et al, 1977) little interaction with Kupifer cells was observed. In the present investigation, however, we have seen apparently intact TA3 cells, particularly TA3/Ha cells, that were being surrounded by Kupffer cell processes.…”

“…Perfusion technique.-The perfusion procedure has been described previously (Roos et al, 1977). Briefly, mouse livers were perfused in situ through the vena porta with an oxygenated Krebs-Ringer buffer supplemented with glucose, albumin and amino acids, without an oxygen carrier.…”

“…1,2 Intrahepatic trafficking and arrest of circulating cancer cells constitutes a critical step in hepatic metastasis development whose functional regulation is far from being clear. The process involves an initial trapping of cancer cells in the proximal segment of sinusoidal pathways, 3 which leads to a significant cancer cell destruction caused by mechanical stress. 4,5 Next, a reciprocal functional interaction occurs between surviving cancer cells and sinusoidal cells, leading to an acute inflammatory response of the latter cells.…”
