Investigation of TiO2nanoparticles translocation through a Caco-2 monolayer

Abstract: Abstract. Nanoparticles (NPs) are introduced in a growing number of commercial products, including food and beverage but their effects on gastrointestinal tract are poorly investigated. Here we focused on the translocation of TiO 2 NPs through Caco-2 monolayers exposed to anatase and rutile NPs up to 24 h. Internalization was followed by transmission electronic microscopy and µ-XRF elemental mapping, coupled to XAS analysis of Ti atoms environment. This innovative technique is among the best techniques to get … Show more

“…Brun et al studied this translocation using transmission electron microscopy and synchroton-based micro X-ray uorescence. 38 in the sample collected from the basolateral pole, indicating that translocation of the TiO 2 NPs appears to be minimal. In fact, the number of signal pulses was not signicantly different from blank control samples in the experiment and in that case the translocation was quantied by considering the number of signal pulses plus three times the standard deviation in the number of signal pulses in blank samples as the detection limit, LOD CONC .…”

“…used cultured monolayer assays to show that nanosized pharmaceuticals have higher bioavailability than larger‐sized pharmaceuticals (Jia ). Using Caco‐2 cell monolayers to investigate TiO 2 nanoparticle translocation across epithelial cells, Brun and others () coupled in vitro translocation studies with transmission electron microscopy (TEM) to show that the TiO 2 nanoparticles accumulated in the cells. Other studies that have used Caco‐2 transport models to investigate ENM uptake or interactions with intestinal cells have also been published (Jia ; Lin and others , ; Jia and others ; Zhang and others ).…”

“…Figure shows that in the case of the reference CaPs analyzed (ACCP, CA, and HAP), the prepeak is quite similar but it is not the case for all compounds. An educational case is given with TiO 2 , for which the observed pre-edge features vary both in positions (by ∼2 ± 0.1 eV) and normalized height (from ∼0.04 to 1.0 ± 0.05) as a function of Ti coordination (4, 5, or 6 oxygen nearest neighbors). …”
