Is Liver Enzyme Release Really Associated with Cell Necrosis Induced by Oxidant Stress?

Abstract: Hepatic diseases are a major concern worldwide. Increased specific plasma enzyme activities are considered diagnostic features for liver diseases, since enzymes are released into the blood compartment following the deterioration of the organ. Release of liver mitochondrial enzymes is considered strong evidence for hepatic necrosis, which is associated with an increased production of ROS, often leading to greater hepatic lipid peroxidation. Lipotoxic mediators and intracellular signals activated Kupffer cells, … Show more

“…However, some are more commonly used; the ALT and AST enzymes are indicated for morphological analysis of the liver (hepatocytes), and the enzymes ALP and GGT to check for cholestasis (Opara et al 2006, Contreras-Zentella & Hernández-Muñoz 2016. The mean ALT and AST levels found in this study were lower than those previously reported for the species, while the GGT levels were higher.…”

“…It is one of the most important components of human diet due to its ability to mop-up and inhibits further generation of ROS which accounts for its several pharmacological activities (Laight, 2000). Elevated transaminase, such as ALT and AST, activities are known marker for hepatocyte injury mediated by ROS via oxidative stress (Contreras-Zentella and Hernández-Muñoz, 2016). GGT is a specific biomarker of hepatobiliary injury, especially cholestasis and biliary effects (Sheehan and Haythorn, 1979), while ALP is a marker enzyme for the plasma membrane and endoplasmic reticulum (Wright and Plummer, 1974).…”

“…The latter (cytosolic fraction) could be part of a signaling pathway activating STATs translocation into the nucleus, as well as stimulating cyclin D1 expression [10, 11]. In fact, we have demonstrated that increased serum levels of cytoplasmic enzymes observed during PH-induced rat liver regeneration is differentially regulated by modifications of the oxidant status, indicating that this release is a strictly controlled event [54]. …”
