This study shows differences at the level of cell cycle arrest between the response of yeast cells to hydrogen peroxide and superoxide stress. These include both cell cycle phases at which arrest occurs and the involvement of the RAD9 checkpoint gene. Wild-type and rad9 cells were treated with hydrogen peroxide or the superoxidegenerating agent menadione. rad9 mutants were up to 100-fold more sensitive to hydrogen peroxide but not affected in their resistance to menadione. Hydrogen peroxide caused G 2 -phase arrest, whereas menadionetreated cells arrested in G 1 . G 2 arrest, induced by methyl 2-benzimidazil carbamate, increased cellular resistance to hydrogen peroxide but not to menadione. G 1 arrest mediated by ␣-factor caused an increase in survival of wild-type cells treated with menadione but not with hydrogen peroxide. A cdc28 mutant arrested in G 1 was significantly more sensitive to hydrogen peroxide than other cdc mutants arrested in later phases, including G 2 . rad9 cells have normal stationary phase resistance to hydrogen peroxide, the ability to adapt to it, glutathione content and induction of genes via the stress responsive element. Although rad9-dependent G 2 arrest is important, other rad9-dependent factors may be involved in the resistance of cells to hydrogen peroxide since arrest in G 2 did not make rad9 cells fully resistant.

Mutations in the Saccharomyces cerevisiae alpha-factor receptor that lead to improved response to Saccharomyces kluyveri alpha-factor were identified and sequenced. Mutants were isolated from cells bearing randomly mutagenized receptor gene (STE2) plasmids by an in vivo screen. Five mutations lead to substitutions in hydrophobic segments in the core of the receptor (M54I, S145L, S145L-S219L, A229V, L255S-S288P). Remarkably, strains expressing these mutant receptors exhibited positive pheromone responses to desTrp1,Ala3-alpha-factor, an analog that normally blocks these responses. The M54I mutation appeared to affect only ligand specificity. The other mutations conferred additional effects on signaling or recovery. Two mutants were more sensitive to alpha-factor than wild type (S145L, A229V). One mutant was more sensitive to alpha-factor-induced cell cycle arrest initially, but then recovered more efficiently (S145L-S219L). One mutant (L255S-S288P) conferred positive pheromone responses to alpha-factor as assayed by FUS1-lacZ reporter induction, but did not display growth arrest. The hydrophobic receptor core thus appears to control activation by some ligands and to play roles in aspects of signal transduction and recovery.

A peptide, termed CI" pheromone, was isolated as a mating pheromone from culture filtrate of mating type a cells of Saccharomyces exiguus. The peptide showed both agglutinability-inducing activity to a cells of S. cerevisiae and shmoo-inducing action to a cells of S. cerevisiae, S. kluyveri and S. exiguus. The amino acid sequence of use pheromone was determined as H-Trp-His-Trp-Leu-Arg-Leu-Ser-Tyr-Gly-Gln-Pro-IleTyr-OH by mass spectrometry, sequence analysis and enzymatic digestion.
