Isolation and amino acid sequence of a mating pheromone produced by mating type α cells of Saccharomyces kluyveri

Abstract: A peptide, termed ask2 pheromone, was isolated from culture filtrates of mating type a cells of Saccharomyces kluyveri as a mating pheromone, having both shmoo inducing (in a cells of S. kluyveri and S. cerevisiae) and agglutinability inducing (in a cells of S. cerevisiae) actions. The amino acid sequence of Ly sk2 pheromone was determined as H-Trp-His-Trp-Leu-Ser-Phe-Ser-Lys-Gly-Glu-Pro-Met-Tyr-OH by mass spectrometry, sequence analysis and enzymatic digestions. “…Wild-type and rad9 cells in exponential phase (A 600 of 0.5) were treated with either ␣-factor or MBC, which arrest cells in G 1 and G 2 /M phase, respectively (26,27). Cultures were incubated for a further generation time (3 h) and arrest was confirmed by microscopic observation before treatment with various concentrations of either hydrogen peroxide or menadione.…”

“…The budding yeast Saccharomyces kluyveri, only distantly related to S. cerevisiae, makes an a-factor peptide (k-a-f) that, like c-a-f, is 13 amino acid residues long but that differs in sequence at five positions (9,34). The k-a-f receptor (k-STE2) has been cloned and sequenced and is 50% identical to STE2 on an amino acid level (23).…”

“…Since the other two pheromones, Ly SC pheromones and ask pheromones, showed cross-action between a cells of S. cerevisiae and S. kluyveri [3], these three pheromones have the same biological action. use pheromone obtained in this study appears to be different in amino acid composition, since it contains arginine and isoleucine instead of lysine and methionine which are usually contained in the other two pheromones.…”

“…Amino acid compositions were determined by acid hydrolysis of each peptide (2-5 nM) with 5.7 N HCl containing 3% thioglycolic acid in an evacuated tube at 110°C for 20 h, followed by amino acid analysis on a Hitachi 835 amino acid analyzer. Mass spectrometric analysis by SIMS was perpheromone, which was obtained in a yield of formed on a Hitachi M80 spectrometer as in [3].…”
