Isolation and Characterization of a Novel Noncoding RNA from Nickel-Induced Lung Cancer

Abstract: Noncoding RNAs have drawn significant attention in carcinogenesis. In this study, we identified a novel gene named nickel-related gene1 (NRG1) associated with nickel-induced cancer. By using rapid amplification of cDNA end PCR, we obtained the full length of the cDNA. The sequence was analyzed by using related bioinformatics software and comparative genomics methods. The results showed that NRG1 was located on chromosome 2q12, within intron2 of ADAMTS6, a disintegrin and metalloproteinase with thrombospondin m… Show more

“…Nickel toxicities include skin allergies, lung fibrosis, and lung cancer (Zhao et al, 2009). A recent study has identified a novel 1280 bp non-coding RNA NRG1 associated with nickel-induced lung cancer and has suggested the potential role of lncRNAs in nickel carcinogenesis (Zhang et al, 2012).…”

“…The full-length cDNA of CRNG was obtained via RACE. Simultaneously, for identifying the characteristic of CRNG, genome location and coding potential were analyzed via NCBI Genome and ORF Finder (Zhang et al, 2012), CPC (Kong et al, 2007), respectively. Additionally, the ORF finder can search novel DNA sequence for potential protein encoding fragments, then verify predicted protein via SMART BLAST or regular BLASTP.…”

“…Many lncRNAs have been reported to be associated with non-small cell lung cancer (NSCLC), such as CAR10, MALAT1, HOTAIR and so on [32][33][34]. Studies have shown that lncRNA was also involved in nickel-induced tumorigenesis [35,36]. A new lncRNA, termed nickel-related gene (NRG1), was the first to be identified as a lncRNA induced in Ni-induced lung cancer but the mechanisms that regulate nickel-induced NRG1 level is not known [37].…”
