Isolation and characterization of a thermostable cellulase-producing Fusarium chlamydosporum

Abstract: A thermostable cellulase-producing fungus, HML 0278, was identified as Fusarium chlamydosporum by morphological characteristics and ITS rDNA sequence analysis. HML 0278 produced extracellular cellulases in solid-state fermentation using sugar cane bassage as the carbon source. Native-PAGE analysis demonstrated that this fungus strain was capable of producing the three major components of cellulases and xylanase, with a yield of 281.8 IU/g for CMCase, 182.4 IU/g for cellobiohydrolase, 135.2 IU/g for b-glucosida… Show more

“…For examples, Kwon et al used esculin to visualize β-glucosidase activity on Native-PAGE. The gel was removed and soaked in solution containing 0.1% esculin and 0.03% ferric ammonium citrate until a dark band appeared on the gel indicating the presence of β-glucosidase activity [139,166,167]. P-PNG has also been utilized to detect β-glucosidase activity on PAGE.…”

“…And the results that maximum cellulase activities occurred at different points in different substrates, were probably due to the different expression of enzymes, which might influence the synergistic action towards the insoluble cellulose. Thus, these strains produced all three required components of the cellulase enzyme complex, which hydrolyzed crystalline cellulose into oligosaccharides first and ultimately into glucose (Qin et al 2010). Currently, most commercial cellulases are produced by Trichoderma species and Aspergillus species ).…”

“…3B, which was similar to cellobiohydrolases from other thermophilic fungi, such as Fusarium chlamydosporum, Thermoascus aurantiacus and Thielavia terrestris. (Qin et al, 2010;Hong et al, 2003;Woon et al, 2016). The bivalent metallic cations Mg 2+ and Ca 2+ significantly enhanced the activity of CtCel6, while the monovalent cations K + and Na + were not detected with obvious effect (Table 1).…”
