Isolation and Characterization of a Protective Antigen-Containing Particle from Culture Supernatant Fluids of Erysipelothrix rhusiopathiae

Abstract: The mouse-protective activity of Erysipelothrix rhusiopathiae culture supernatant fluids exists in a polydisperse form, ranging in density from aggregates which sediment at 10,000 × g for 3 hr to soluble units which will not sediment at 198,000 × g for 12 hr. A partially purified protective antigen has been isolated from the aggregates sedimented from a concentrate of the culture supernatant fluid at 20,000 × g for … Show more

“…It has been reported that the protective antigen of E. rhusiopathiae is also present in the culture supernatant of the organism and that antiserum against the culture supernatant fluids is protective (8,9,15,16). Although we found that SpaA.1 is also present in the culture supernatant, we do not yet know the relationship between the bound SpaA.…”

“…From these results it was suggested that the pigs having the specific antibody to P64 could resist infection with the virulent strain (Fujisawa) of E. rhusiopathiae and that P64 is a cross-protective antigen, as P64 from strain Agata (serovar 5) could protect pigs against the infection caused by strain Fujisawa (serovar 1a). Such a cross-protective antigen of E. rhusiopathiae has been reported by many investigators (WHITE and VERWEY, 1970;SAWADA and TAKAHASHI, 1987a, b;SATO et al, 1995).…”

“…The accompanying paper (7) reported on the isolation and characterization of a large protective antigen-containing particle or aggregate found in culture supernatant fluid of Erysipelothrix rhusiopathiae. The protective antigen present in these particles was representative of the major protective antigen of E. rhusiopathiae in that it was capable of absorbing all of the protective activity present in anti-whole culture rabbit serum.…”
