Isolation and Characterization of a γ-Thiochromanone-4-Thiosemicarbazone-Resistant Mutant of Vaccinia Virus

Abstract: γ-Thiochromanone-4-thiosemicarbazone (TCT) inhibits the growth of vaccinia virus in BSCl cells by interfering with viral maturation. A mutant of the virus (TCT R ) which is resistant to this drug was isolated. This mutant also exhibits resistance to another thiosemicarbazone related compound, isatin β-thiosemicarbazone (IBT). There is a good correlation between the cross-resistance of the two mutants IBT R and TCT R to TCT and IBT, respect… Show more

“…No additive effect of the two compounds was observed in the animal tests. The thiochromanone-TSC activity in tissue culture has been confirmed (KATZ et al 1974(KATZ et al , 1975. A single report (Buu-HOI et al 1968) exists indicating that 3,9-diethyl-6-formylcarbazole-TSC (Fig.…”

“…21) was shown to inhibit vaccinia virus replication beyond the adsorption-penetration-uncoating stage because it could be effectively added to chick embryo cell cultures up to 8 h after infection (TSUNODA et al 1971). KATZ et al (1975) using techniques developed in studies on the mode of action of IBT against vaccinia virus in HeLa cells (Sect. F.III) found that TCT did not inhibit the replication of vaccinia DNA.…”

“…Thiosemicarbazones appear to be a structural class with anti-pox virus activity (Katz, 1987;Katz et al, 1975;Rao et al, 1966). Methisazone (I) plays an important role as prophylactic agent against several viral diseases (Sethi, 2002).…”
