Isolation and characterization of calmodulin from an insulin-secreting tumour

Abstract: A major protein constituent of a rat islet cell tumour that exhibited Ca2+-dependent changes in electrophoretic mobility has been purified to homogeneity and compared in its physicochemical and biological properties with bovine brain and rat brain calmodulin (synonymous with phosphodiesterase activator protein, calcium-dependent regulator, troponin C-like protein and modulator protein). The protein, like these calmodulins, contained trimethyl-lysine, exhibited a blocked N-terminus and had an identical amino-ac… Show more

“…Tissues. Insulinomas were propagated in New England Deaconess Hospital (NEDH) rats as described previously [14]. Tumours were removed and tissue was scraped from the fibrous capsules into Hanks saline [(in retool/l) 137 NaC1, 5.4 KC1, 1.67 MgSO 4, 4 CaC12, 0.34 NazHPO 4 and 4.2 NaHCO 3 (pH 7.4)].…”

“…Samples (2-50 c(g protein) were incubated for I-20 mm at 30°C in 50 /.d 50 mM MES (pH 6.9) containing 1 mM MgC12, 0.1 mM DTT, 5.1 mM EGTA and 60 PM [Y-~~P] ATP (4 X lo6 dpm; prepared as in [21] from carrier free 32Pi; Amersham Intl., Bucks.). Other additions included 5 /.IM chicken gizzard MLC (prepared as in [22]), 1.2 PM bovine brain calmodulin (prepared as in [9]) and 5 mM CaC12 which resulted in free Ca2+ estimated at 15 PM [23]. The reaction was terminated by addition of 30 /.d 200 mM TrisHCl (pH 6.8) containing 20% (w/v) sucrose, 2% (w/v) SDS, 10 mM DTT, 0.02% bromophenol blue and heating to 1OO'C for 5 min.…”
