Isolation and characterization of human cervical-mucus glycoproteins

Abstract: Mucus glycoproteins (mucins) were extracted from human cervical pregnancy mucus by 6 M-guanidinium chloride in the presence of proteinase inhibitors. Purification was subsequently achieved by isopycnic density-gradient centrifugation in CsCl/ guanidinium chloride gradients. The purified macromolecules represented approx. 85% of the total and were devoid of nucleic acids and proteins, as judged by analytical density-gradient centrifugation, disc electrophoresis and u.v. spectroscopy. Sedimentation-velocity cent… Show more

“…4). While proteins and glycoproteins in general have densities not exceeding 1.3 g/ml, this value is in agreement with the known characteristics of mucus glycoproteins [17]. A similar buoyant density has been reported for a Ca19-9 active serum mucin [18].…”

“…At the trans‐golgi compartment, the addition of sialic acid or GalNAc will cease glycan extension 31. The end result is a highly decorated protein dimer with nearly 80% of the mass accounted for by glycans 32. These glycan chains are highly heterogeneous in chain length and composition even at homeostasis.…”

“…Other groups have included 4 -6 M guanidinium hydrochloride in the extraction solution and in the CsCl gradient [15,16]. The problem with this is that it is a chaotropic agent and will denature the mucins and reduce their ability to form a gel.…”
