Isolation and characterization of lectin from the leaves of Euphorbia tithymaloides (L.)

Abstract: Lectins or glycoproteins from the leaves of Euphorbia tithymalo were isolated after screening of different flora from Central India. The crude extract of leaves was dialyzed andammonium sulfate precipitation done and followed by dialysis for the purification of lectins. Protein concentration in the purified extract was 10 mg.ml -1 measured by Biuret Method. The purified lectins were able to agglutinate human erythrocytes of ABO blood group system. Agglutination was also visible with animal erythrocytes. Lectin… Show more

“…It is known that, phytolectins have property to specifically bind glycoconjugates on the cell surface and agglutinates RBCs of human as well as animals. Plant extract also agglutinates some of the animal erythrocytes which is supported by study of Euphorbia tithymaloids by Jawade et al (2016), Ahmed & Cahtterjee (1987) and with Tridax procumbans by Ramteke & Patil (2005).…”

“…The solution was incubated for 1 h at room temperature and observed visually. Positive results of the agglutination reaction were indicated by a uniform red color in the solution, while negative results showed a red dot precipitate [18].…”

“…The electrophoresis procedure refers to Jiang et al [17] and Jawade et al [18] with slight modifications. Crude extract samples, A1, B1, B2, C1, and C2, were mixed with Coomassie brilliant blue loading dye in a ratio of 5:1 and heated in a water bath at 95-100℃ for 5 min.…”

“…It is also crucial to note the activity of proteins present in the Euphorbiaceae latex. Euphorbia trigona latex is high in lectin-content, which is responsible for precipitation and agglutination in cells through binding specifically and reversibly to carbohydrates ( Jawade et al., 2016 ). Three of those lectins have been characterized and their RNA N-glycosidase activity was tested on several human cancer cell lines, including HeLa, A549, HCT116, HL-60, human colorectal HT-29.…”
