Isolation and Characterization of Probiotic Bacillus subtilis MKHJ 1-1 Possessing L-Asparaginase Activity

Abstract: The purpose of this study was to isolate functional Bacillus strains from Korean fermented soybeans and to evaluate their potential as probiotics. The L-asparaginase activity of MKHJ 1-1 was the highest among 162 Bacillus strains. This strain showed nonhemolysis and did not produce β-glucuronidase. Among the nine target bacteria, MKHJ 1-1 inhibited the growth of Escherichia coli, Pseudomonas aeruginosa, Shigella sonnei, Shigella flexneri, Klebsiella pneumoniae, Staphylococcus aureus, and Bacillus cereus. 16S r… Show more

“…Except for OM02 and OM04, all other strains were found to be non-β-haemolytic, and making them suitable for usage as probiotics. Our findings are supported by research that has looked at and documented the β-haemolytic character of some strains of Bacillus cereus (24), γ-haemolysis in Bacillus velezensis GPSAK4, and Bacillus subtilis GPSAK9 (21). Furthermore, the antibiotic susceptibility study illustrated that OM03 and OM05 are susceptible to all the six antibiotics tested, as they did not harbour any of the antibiotic-resistant genes against the tested.…”

“…2). Previous research has shown that the Lasparaginase activity of the probiotic Bacillus subtilis strain derived from fermented soybeans in Korea was 1.20 U/mL (24). According to the findings of another investigation, L-asparaginase obtained from Lactobacillus sp.…”

“…LAB belonging to the Enterococcus, Fructobacillus, Lactobacillus, Lactococcus, Leuconostoc, and Weissella genera were isolated from fruits of papaya, guava (pink and yellow varieties), custard apple, passion fruit, medlar, mulberry, khaki, and fig, and flowers of passion fruit, papaya medlar, and custard apple, in MRS medium [17]. Bacillus strains were isolated from Korean fermented soybean [58]. Lactobacilli were isolated from the oral cavity, dairy products, and feces using MRS agar at 28 • C [59].…”

“…Disposal of effluents by industries and mismanagement of sewage waste are the two major issues that hamper the quality of drinking water. 8,9 All effluents can either be released into a stream without any treatment or only after the necessary treatment, regulations regarding sanitary conditions and pollution levels may differ.
