Isolation and expression analysis of a novel pathogenesis-related protein 10 gene from Chinese wild Vitis pseudoreticulata induced by Uncinula necator

Abstract: A novel PR10 gene (designated as VpPR10) was isolated from Chinese wild Vitis pseudoreticulata W. T. Wang Baihe-35-1. VpPR10 encoded a 159-amino-acid polypeptide with a predicted molecular mass of 17.46 kDa. Sequence alignment showed that VpPR10 had high similarity with the PR10 proteins of other plant species. Genomic DNA gel blot analysis indicated that VpPR10 belonged to a multi-gene family in Chinese wild Vitis. Quantitative real-time PCR analysis exhibited that the transcript level of VpPR10 altered at di… Show more

“…The Ypr6 (proteinase inhibitor), Ypr12 (plant defensin), Ypr13 (plant thionin), and Ypr14 (lipid transfer protein) antimicrobial peptides are usually cysteine-rich peptides with broad-spectrum antifungal activities ( Loon et al, 2006 ; Nawrot et al, 2013 ). Many PR10 proteins and PR4 protrins have been shown to have RNase activity ( Xu et al, 2010 ; Wang et al, 2011 ; Yang et al, 2018 ). The recombinant Theobroma cacao PR-10 protein has antifungal activities against the basidiomycetous pathogen Moniliophthora perniciosa and the yeast Saccharomyces cerevisiae , while also exhibiting in vitro and in vivo RNase activities ( Pungartnik et al, 2009 ).…”

“…These families have been numbered in sequence of discovery ( Sels et al, 2008 ; Sinha et al, 2014 ). Usually, PRs possess enzymatic functions – so, for example, PR-2 is a 1,3-β-glucanase ( Kauffmann et al, 1987 ), PR-7 has endoproteinase activity, PR-9 is a peroxidase, and PR-10 is a ribonuclease ( Lagrimini et al, 1987 ; Vera and Conejero, 1988 ; Park et al, 2004 ; Xu et al, 2010 ). Moreover, a number of them, e.g., PR-1 and PR-6, have been identified as having antifungal and proteinase inhibitory properties ( Green and Ryan, 1972 ; Niderman et al, 1995 ).…”

“…Since 1999, in different genetic backgrounds several genes that may contribute to PM resistance have been identified, such as VpGLOX (Guan et al, 2011); VpPR-10.1 (Xu et al, 2010b; Xu et al, 2014); VpRFP1 (Yu et al, 2011); VpALDH2B4 (Wen et al, 2012); VpNAC1 (Zhu et al, 2012b); VpPR4-1 (Dai et al, 2016) ( Tables 2A and S2A). Many of them are transcription factors (Zhu et al, 2012a; Zhu et al, 2012b), as expected since they are master regulators in controlling plant response to biotic stress (Amorim et al, 2017; Ng et al, 2018).…”
