Isolation and Structures of Two New Sesquiterpenoid Aromatic Esters: Armillarigin and Armillarikin1

Abstract: The continued investigation of the mycelial extract of ARMILLARIA MELLEA (Vahl.ex Fr.) Quel. (Tricholomelaceae) led to the isolation of two new sesquiterpenoid aromatic esters, named armillarigin ( 1) and armillarikin ( 2). Their structures were elucidated on the basis of spectral data and chemical transformation.

“…The armillarikin was purified as previously described. 2 , 9 , 18 In brief, the ethanol (EtOH) extract of A. mellea was separated by using silica gel column chromatography, which was eluted with n -hexane:ethyl acetate (EtOAc, 1:1). The final extract obtained was dissolved in dimethyl sulfoxide (DMSO, Sigma-Aldrich Co., St Louis, MO, USA) as a 1,000-fold stock solution.…”

“…Three fractions were obtained with 20% diethyl ether-petroleum ether elution, and the third fraction (0.85 g) was rechromatographed on a column of silica gel (160 g) and eluted with henzene-diethyl ether (9: 1) to give 17 fractions. The 11-12th fraction afforded a colourless oil, named armillatin (2) (100 mg).…”

“…(Tricholomataceae) resulted in the isolation of a number of sesquiterpenoid aromatic esters possessing the protoilludane skeleton. Our further investigation of the artificially cultured mycelium of A. mellea has now led to the isolation of two additional novel protoilludane norsesquiternoid aromatic esters named armillasin (1) and armillatin (2). The present paper deals with their isolation and characterization.…”
