Isolation, characterization and expression of a gene coding for a 2S albumin from Bertholletia excelsa (Brazil nut)

Abstract: Two genes, BE2S1 and BE2S2, coding for methionine-rich albumins of Brazil nut (Bertholletia excelsa H.B.K.) have been cloned and their sequence determined. The genes are members of a multigene family and one of them, i.e. BE2S1, codes for one of the dominant 2S isoforms. Its expression is highly regulated during seed development and with respect to tissue specificity. Sequence analysis has shown that the genes contain one intron and that the promoter of BE2S1 shows a canonical TATA motif. The transcription ini… Show more

“…A detection level of 70% or more in conjunction with a minimal false alarm rate was judged as optimal. Accordingly, this setting was chosen for specific tests on the Brazil nut 2S albumin and the modified Cry9C protein [40, 41, 42]. The query procedure included the 2S albumin amino acid sequence in the test data set in 44 of the 200 sampling cycles.…”

“…Plants were co-transformed with plasmids pEA23 and pBI426 (circular form). Plasmid pEA23 contains the β-glucuronidase (GUS) coding region (uidA gene) under control of the 35S CaMV promoter and the 2S-albumin gene from the Brazil nut (be2s2) (Gander et al, 1991), driven by the doubled 35S CaMV promoter plus the AMV leader sequence (Datla et al, 1993). Plasmid pBI426 containing the uidA-neo gene fusion (Datla et al, 1991), driven by the doubled 35S CaMV promoter plus an enhancer sequence from AMV, was provided by Dr. William Crosby (Plant Biotechnology Institute, Saskatoon, Canada).…”

“…We have isolated and characterized one of the Brazil nut's 2S-albumin genes (be2s1 gene) (Gander et al, 1991). This gene codes for an 18-kDa precursor protein, which is processed by a complex series of steps to yield two subunits of 3 and 9 kDa, joined by disulfide bridges (Sun et al, 1987).…”

“…The 2S albumins are products of multigene families, and therefore various isoforms may exist. Also for Ber e 1 a number of isoforms have been identified [17][18][19]. Various procedures for the purification of Ber e 1 have been described [14,15,[21][22][23], but most of these methods consist of several time-consuming centrifugation and/or column chromatography steps, making them only suitable for the purification of small quantities of Ber e 1.…”
