Isolation, characterization and genetic diversity of NBS-LRR class disease-resistant gene analogs in multiple virus resistant line of chilli (Capsicum annuum L.)

Abstract: Viruses are serious threat to chilli crop production worldwide. Resistance screening against several viruses resulted in identifying a multiple virus resistant genotype 'IHR 2451'. Degenerate primers based on the conserved regions between P-Loop and GLPL of Resistance genes (R-genes) were used to amplify nucleotide binding sites (NBS)-encoding regions from genotype 'IHR 2451'. Alignment of deduced amino acid sequences and phylogenetic analyses of isolated sequences distinguished into two groups representing to… Show more

“…We identified a total of 571 NBS-ARC domain genes, 390 of which had a complete NBS-ARC domain. Before our study, only dozens of NBS-ARC genes were identified by degenerate PCR amplification [ 15 , 16 ]. This is due to either the limitation of degenerate PCR or the absence of genomic sequences at that time.…”

“…NBS-ARC-LRR protein genes constitute the predominant majority of disease resistance genes ( R genes). Several doses of R genes have been amplified in peppers by degenerate primers [ 15 , 16 ]. However, most R genes are still unknown because higher plants typically have hundreds of R genes.…”

“…Yadav et al [169] also identified the LRR LIKE-PROTEIN KINASE gene as MYMIV-resistance in soybean. Likewise, Naresh et al [170] characterized the NBS-LRR class disease resistance gene analogs leading to multi-virus resistance in chilli, indicating the need for deep analysis of NBS-LLR candidates for YMD-resistance in mungbean. These candidates will also help in developing gene-specific markers for resistance breeding.…”

“…These primers are designed based on sequences of DNA resistance genes from other plants, known as resistance gene analogs (RGA) (Naresh et al 2017). One of the largest RGA classes that have been characterized is the Nucleotide Binding Site.…”
