Isolation Methods of Peripheral Blood Mononuclear Cells in Spanish Biobanks: An Overview

Abstract: The Spanish Hematic Derivatives Group, consisting of 26 biobanks, was established in 2011. We describe here the viability results of our publically available standard operating procedure to freeze and thaw peripheral blood mononuclear cells (PBMCs). Our protocol maximizes PBMC viability while avoiding where possible interbiobank and intrabiobank assay variability.

“…Human peripheral blood mononuclear cells (PBMCs) were isolated from buffy coats obtained from anonymized healthy blood donors coming from the transfusion Center of Madrid following national guidelines. PBMCs were isolated by a standard Ficoll - Hypaque density gradient (Rafer, Madrid, Spain) and cultured following the procedures of Spanish HIV HGM BioBank [4245]. PBMCs were kept with 60 U/mL of interleukin 2 (IL-2; Bachem AG, Bubendorf, Switzerland) and stimulated with 60 U/mL of interleukin 2 (IL-2; Bachem AG, Bubendorf, Switzerland) and 2 μg/mL of phytohemaglutinin (PHA; Remel, Santa Fe, NM, USA) for 48 h before the experiments for T cell analysis.…”

PBMCs were isolated by a standard Ficoll-Hypaque density gradient (Rafer, Madrid, Spain) and cultured following the procedures of Spanish HIV HGM BioBank [ 21 , 22 , 23 ]. PBMCs were cultured in RPMI-1640 medium (Biochrom AG, Berlin, Germany) with 10% fetal bovine serum (FBS; Gibco, London, UK), 1% l -glutamine (Lonza, Walkersville, MD, USA) and antibiotic cocktail (125 mg/mL ampicillin,125 mg/mL cloxacillin and 40 mg/mL gentamicin; all from Normon, Madrid, Spain).…”

“…PBMC were then immediately processed for flow cytometry or frozen in fetal bovine serum/7.5% dimethyl sulfoxide (DMSO) (Sigma Aldrich) (Consuegra et al, 2017;Mitrus et al, 2013) and conserved in liquid nitrogen for 3 months. For whole blood freezing, samples were precooled 15 min on ice.…”
