The photorespiratory pathway is comprised of enzymes localized within three distinct cellular compartments: chloroplasts, peroxisomes, and mitochondria. Photorespiratory enzymes are encoded by nuclear genes, translated in the cytosol, and targeted into these distinct subcellular compartments. One likely means by which to regulate the expression of the genes encoding photorespiratory enzymes is coordinated temporal control. We have previously shown in Arabidopsis that a circadian clock regulates the expression of the nuclear genes encoding both chloroplastic (Rubisco small subunit and Rubisco activase) and peroxisomal (catalase) components of the photorespiratory pathway. To determine whether a circadian clock also regulates the expression of genes encoding mitochondrial components of the photorespiratory pathway, we characterized a family of Arabidopsis serine hydroxymethyltransferase (SHM) genes. We examined mRNA accumulation for two of these family members, including one probable photorespiratory gene (SHM1) and a second gene expressed maximally in roots (SHM4), and show that both exhibit circadian oscillations in mRNA abundance that are in phase with those described for other photorespiratory genes. In addition, we show that SHM1 mRNA accumulates in light-grown seedlings, although this response is probably an indirect consequence of the induction of photosynthesis and photorespiration by illumination.

Telomerase is a ribonucleoprotein enzyme which elongates the G-rich strand of telomeric DNA to compensate for the progressive reduction in its length due to incomplete replication of chromosome ends, which in human somatic cells leads to cell cycle arrest upon shortening of telomeres to a critical length. To examine the possible involvement of telomerase in metabolism of plant genetic material, we used cells of Nicotiana tabacum strain TBY-2, a stable long-term culture which has kept a constant pattern of restriction fragments from chromosome termini during its 6 month period of cultivation in our laboratory. In a direct assay for telomerase, a 5' end-labeled plant telomeric oligonueleotide S'(TTTAGGG)6 3' was elongated in a TBY-2 cell extract, showing a pausing pattern which is a characteristic feature of telomerases from other organisms. The elongation was inhibited by RNase A pretreatment of the extract. We conclude that plant cells possess telomerase which is used for maintenance of their telomeres.

McClintock's analysis of chromosome-breaking Dissociation (Ds) elements in maize demonstrated that sister chromatids fuse at the position of Ds, forming a dicentric chromosome and an acentric fragment. In tobacco, Ds left and right ends e in direct orientation (that is, half a double Ds) are sufficient to promote Ac!ivator-dependent marker gene loss. We present here a detailed analysis of germinally inherited rearrangements promoted by "half double Ds" elements and a characterization of rearrangements that involve inversion of the segment between the Ds ends and/or deletion of a segment adjacent to the Ds construct. The results support a model in which chromosome breakage promoted by these elements, and presumably by double Ds elements, involves Ds ends on sister chromatids.
