Isolation of Circular Deoxyribonucleic Acid from Salmonella typhosa Hybrids Obtained from Matings with Escherichia coli Hfr Donors

Abstract: Heterozygous, partial diploid Salmonella typhosa hybrids obtained from matings with Escherichia coli K-12 Hfr strains were observed to contain supercoiled, circular deoxyribonucleic acid (DNA) when examined by the dye-buoyant density method. Examination of one such S. typhosa hybrid after its loss, by segregation, of the inherited E. coli genetic markers revealed a concurrent loss of its supercoiled circular DNA. Subsequent remating of this segregant with various E. coli Hfr strains resulted in the reappearanc… Show more

“…As described previously (5), the plaques formed by OH on F-containing (male) E. coli K-12 strains are significantly smaller than those formed on non-F-containing (female) K-12 strains. Experiments to detect increases in phage titer were performed by adding phage to bacteria at a ratio of 1 to 1,000 in appropriate nutrient broth medium, incubating overnight, and assaying the supematants on sensitive indicator bacteria, as described previously (2,7). RESULTS Transfer of the scr elements to E. coli.…”

“…The entire lysate was centrifuged in an ethidium bromidecesium chloride (EtBr-CsCl) density gradient at 44,000 rev/min for 40 hr. Details of the method have been described by Leavitt et al (16). After centrifugation, the gradient was fractionated into 0.3-ml fractions, and 50-juliter samples were counted to determine which fractions contained the labeled DNA.…”

“…In a recent attempt (10) to gain some knowledge of the manner in which supernumerary genes are conserved in partially diploid cells, we examined unstable Salmonella typhosa hybrids derived from matings with Escherichia coli K-12 Hfr strains. Some of these S. typhosa hybrids were found to contain supercoiled circular deoxyribonucleic acid (DNA) molecules, whose presence was correlated with the presence of the added E. coli genetic material.…”
