Isolation of expressed sequence tags of skeletal muscle of neonatal healthy and splay leg piglets and mapping by somatic cell hybrid analysis

Abstract: We have isolated 14 differentially displayed and 10 further expressed sequence tags (ESTs) from Musculus biceps femoris of newborn healthy and splay leg piglets. By comparison with EMBL/GenBank data we could identify nine porcine homologues to human genes (TATA box binding protein associated factor B TAF1B; B-cell CLL/lymphoma 7B BCL7B; pyruvate dehydrogenase kinase, isoenzyme 4 PDK4; ribosomal protein S10 RPS10; SPARC-like 1 SPARCL1; epithelial protein lost in neoplasm beta EPLIN; N-myc downstream-regulated g… Show more

“…PDK4 was located at the 7q21.3-q22.1 region in human [6] and 9q12 ± 1/3q21 in swine [21,22]. Alignment between pig chromosome 9 on RH map and human maps on USDA-MARC Pig Database [23], we found some QTLs associated with meat quality and production traits in this region such as body weight at 10 weeks of age (87 cM) [24], average backfat thickness (79.3-90 cM) [25], total saturated fatty acids (61.5-83.3 cM) [26], backfat at tenth rib (96-79 cM) [27].…”

“…Maternal nutrition is undoubtedly a key factor in foetal growth and its subsequent survival which manifests its effects through a complex signalling network that includes IGFs (Hornick et al, 2000;Maak et al, 2001;Wu et al, 2006). However, in the context of foetal fibre number, the precise role of maternal nutrition is not entirely clear.…”

“…The pCDKN3 cDNA was synthesized as described elsewhere [12]. For the amplification of the coding region of the porcine CDKN3 gene, the following primer sets were used: CdknF: 5 GTG AGA CTG CCA GCC ATG AAG3 ; CdknR: 5 TTG ATG ATA GAT GTG CAG CTA ATT T3 ; KAP3F: 5 ATC GAC AGC CTG AGA GAC C3 ; KAP3R: 5 TTG CCA ATA AAA GCT TTA GGA A3 .…”

“…Several biochemical and histo-morphological investigations indicate an immaturity of the skeletal muscle in the affected piglets at birth [1,2,5,14]. We have recently reported the isolation of skeletal muscle EST differentially expressed in M. biceps femoris of healthy and affected piglets [9][10][11][12]. Among the fragments strongly displayed in the splay leg muscle, we identified the porcine CDKN3 (cyclin-dependent kinase inhibitor 3) gene [13].…”
