Isolation of Mycobacterium paratuberculosis from intestinal mucosa and mesenteric lymph nodes of goats by use of selective Dubos medium

Abstract: To isolate Mycobacterium paratuberculosis from contaminated material, a selective medium, selective Dubos medium (SDubos), was developed by supplementing conventional Dubos medium (CDubos) with carbenicillin, polymyxin, trimethoprim, and amphotericin B. The intestine and mesenteric lymph nodes of 1,501 goats were cultured in parallel on SDubos and CDubos after decontamination with oxalic acid. The contamination rate was reduced more than 150 times by the use of SDubos. The number of positive specimens (18, or … Show more

“…Approximately 50,000 goats produce approximately 20 million liters of milk annually, and paratuberculosis occurs endemically in goats in western Norway. The disease is controlled by vaccination, from 1967 to 2001 with a live vaccine (Saxegaard and Fodstad, 1985), and since 2001 with an inactivated vaccine (García Marín et al, 1999). In cattle, clinical paratuberculosis has occurred only sporadically, with no cases since 1979 (Djønne et al, 2001).…”

“…4 JD is a chronic inflammatory disease of ruminants caused by Mycobacterium avium subspecies paratuberculosis (MAP), a slow grower, fastidious, and intracellular pathogen. 5 Chiodini et al 6 isolated cell wall-deficient organisms, also known as spheroplast, from tissue of 6 of 28 (21.4%) patients with CD after long-term incubation culture. The spheroplast cultures then reverted to acid-fast Mycobacterium-like organism after a single in vitro passage.…”

“…Sixty-five tissue fragments of about 5 to 10 g each from 26 patients with clinically active and histologically confirmed M. ulcerans infection were selected. Upon collection, the fragments were placed in 2 ml of modified Dubos medium (11) and transported to the Institute of Tropical Medicine at room temperature.…”
