Isolation of primary microglia from the human post-mortem brain: effects of ante- and post-mortem variables

Abstract: Microglia are key players in the central nervous system in health and disease. Much pioneering research on microglia function has been carried out in vivo with the use of genetic animal models. However, to fully understand the role of microglia in neurological and psychiatric disorders, it is crucial to study primary human microglia from brain donors. We have developed a rapid procedure for the isolation of pure human microglia from autopsy tissue using density gradient centrifugation followed by CD11b-specifi… Show more

“…New insights may be obtained with microglia isolated from human post-mortem brain tissue (Mizee et al, 2017 ), or derived from induced pluripotent stem cells generated from AD patients (Abud et al, 2017 ). Moreover, because neuron-astrocyte-microglia communication plays a crucial role in AD pathogenesis, and microglia activation triggers astrocyte neurotoxicity (Liddelow et al, 2017 ), additional studies using 3D culture models that allow cell-to-cell interplay and best recapitulate AD (Kim et al, 2015 ; Choi et al, 2016 ; Lee et al, 2016 ) should be used to corroborate or complement our findings.…”

“…Tissue was obtained in accordance with the Declaration of Helsinki and the Amsterdam UMC Research Code provided by the Medical Ethics Committee. Human primary peripheral blood mononuclear cells (PBMCs) and T cells were isolated from blood using the Macs Pan T Cell Isolation Kit (Miltenyi Biotec, Bergisch Gladbach, Germany) and human primary microglia was isolated as previously described (35).…”

“…The interphase containing the microglia was transferred to a new tube (the myelin and erythrocyte layers were discarded) and washed twice with DMEM supplemented with 10% FCS, 1% Pen/Strep, 1% gentamycin, and 25 mM HEPES (Invitrogen). Negative selection of granulocytes (previous method only) and positive selection of microglia with anti-CD15-and anti-CD11b-conjugated magnetic microbeads (Miltenyi Biotec), respectively, were performed by magnetic activated cell sorting (MACS) according to the manufacturer's protocol [36]. Brie y, cells were incubated with 10 μl CD15 microbeads for 15 min at 4°C, washed, suspended in bead buffer (0.5% BSA, 2 mM EDTA in PBS, pH 7.2), and transferred to an MS column placed in a magnetic holder.…”
