Isolation of Putative Biosynthetic Intermediates of Prenylated Indole Alkaloids from a Thermophilic Fungus Talaromyces thermophilus

Abstract: The putative key biosynthetic intermediates of prenylated indole alkaloids have long been proposed but never isolated. Two such alkaloids, named talathermophilins A and B (1 and 2), were isolated from a thermophilic fungus Talaromyces thermophilus strain YM1-3 and were identified by NMR and MS spectroscopic analyses. The ratio of 1 and 2 in the culture broths was unexpectedly rather constant (about 2:3), which even remained unchanged despite the addition of exogenous 1 or 2, suggesting that talathermophilins m… Show more

“…), two unusual cyclodepsipeptides containing an anthranilic acid dimer and phenyllactic acid residues were isolated from an A. clavatus strain associated with the thermophilic crab, Xenograpsus testudinatus which lives around the sulphur‐rich hydrothermal vents in Taiwan, these compounds inhibit the proliferation of hepatocellular carcinoma cell lines (HepG2) by arresting their growth at G1 phase (Jiang et al ). Chemical investigation of the thermophilic fungus T. thermophilus afforded two new prenylated alkaloids talathermophilins A and B ( 177 , 178 ) which showed nematicidal toxicity against the parasitic worm Panagrellus redivevus (Chu et al ). Subsequently, four talathermophilins ( 179 – 182 ) were isolated from the same fungus (Guo et al ).…”

“…2 Among distinct types of fungi present in this environment, endophytes are receiving attention due to their ability to produce biologically active natural products. 3,4 Endophytic strains of Talaromyces are still poorly studied, with few works reporting mainly secondary metabolites from the alkaloid, 5 terpene, 6 and polyketide classes. 7 Some of the described compounds possess remarkable pharmacological activities.…”
