Isomer-Specific LC/MS and LC/MS/MS Profiling of the Mouse Serum N-Glycome Revealing a Number of Novel Sialylated N-Glycans

Abstract: Mice are the premier mammalian models for studies of human physiology and disease, bearing extensive biological similarity to humans with far fewer ethical, economic, or logistic complications. To facilitate glycomic studies based on the mouse model, we comprehensively profiled the mouse serum N-glycome using isomer-specific nano-LC/MS and -LC/MS/MS. N-Glycans were identified by accurate mass MS and structurally elucidated by MS/MS. Porous graphitized carbon nano-LC was able to separate out nearly 300 N-linked… Show more

“…Hypersialylation has only recently been described in the bovine (Sumiyoshi et al 2012) and mouse serum (Hua et al 2013). However, similar linkages have previously been described in free oligosaccharides from bovine colostrums (Nemansky & Van den Eijnden 1992), human milk (Sumiyoshi et al 2004) and mucin-type O-linked glycans from human mucosal tissues (Brockhausen 2006).…”

“…The detailed structural information about the aberrant glycans was verified by nanoLC-ESI-MS/MS as mentioned below. Up to now, only a recent article described the appearance of dehydrated sialic acids in the serum of mouse [27]. We supposed that the sialic acid in the glycans were dehydrated and a structure of lactone (NeuAcLac) was formed.…”

“…Graphitised carbon solid phase extraction (SPE) is a well-established glycan purification technique that is commonly used in many glycomics workflows, including characterisation of protein N-glycomes (Aldredge, An, Tang, Waddell, & Lebrilla, 2012; Hua et al, 2013; Stumpo & Reinhold, 2010), biomarker discovery (Alley, Madera, Mechref, & Novotny, 2010; Balog et al, 2012; Vieira et al, 2012), and profiling of free milk OS (Marx et al, 2014; Mudd et al, 2016; Tao et al, 2008). Although previous publications have characterised the excellent retention characteristics of graphitized carbon in the context of glycan desalting and preconcentration (Packer, Lawson, Jardine, & Redmond, 1998; Redmond & Packer, 1999; Ward, 2009), the efficiency of the technique in separating milk OS and lactose has not been well-characterised, despite its widespread use for this purpose.…”
