Context: Fungal infections caused by fluconazole-resistant Candida albicans are an intractable clinical problem, calling for new efficient antifungal drugs. Kaempferol, an active flavonoid, has been considered a potential candidate against Candida species. Objective: This work investigates the resistance reversion of kaempferol in fluconazole-resistant C. albicans and the underlying mechanism. Materials and methods: The antifungal activities of fluconazole and/or kaempferol were assessed by a series of standard procedures including broth microdilution method, checkerboard assay and time-kill (T-K) test in nine clinical strains as well as a standard reference isolate of C. albicans. Subsequently, the morphological changes, the efflux of rhodamine 6G, and the expressions of CDR 1, CDR 2, and MDR 1 were analysed by scanning electron microscope (SEM), inverted fluorescence microscope and quantitative reverse transcription polymerase chain reaction (qRT-PCR) in C. albicans z2003.Results: For all the tested C. albicans strains, the minimum inhibitory concentrations (MICs) of fluconazole and kaempferol ranged 0.25-32 and 128-256 mg/mL with a range of fractional inhibitory concentration index of 0.257-0.531. In C. albicans z2003, the expression of both CDR 1 and CDR 2 were decreased after exposure to kaempferol alone with negligible rhodamine 6G accumulation, while the expression of CDR 1, CDR 2 and MDR 1 were all decreased when fluconazole and kaempferol were used concomitantly with notable fluorescence of rhodamine 6G observed. Discussion and conclusion: Kaempferol-induced reversion in fluconazole-resistant C. albicans might be likely due to the suppression of the expression of CDR1, CDR2 and MDR1.ARTICLE HISTORY

The genus Rhodiola (Crassulaceae) consists of many species, growing mainly in Asia and traditionally used as adaptogens and anti-inflammatory drugs. In order to elaborate herbal immunostimulator which could be safely given to pregnant women, we performed a study on immunotropic effects of feeding pregnant and lactating mice Rhodiola kirilowii extracts. This paper presents the results of the first part of our study -spleen content of selected polyphenols, spleen cellularity, splenocytes phenotype and their response to mitogens. Experiments were performed on adult inbred females of Balb/c strain, mated with adult males. Females, since copulatory plug was noted, up to the 28-th day after delivery were fed daily with 20 mg/kg b.m. water (RKW) or hydro-alcoholic (RKW-A) extracts of Rhodiola kirilowii. Results: 1. Significantly lower proportion of pregnant mice in experimental groups than in the control. 2. Cellularity of spleen and flavonol quercetin spleen concentration were significantly lower in experimental groups in comparison to the controls. 3. Flavanols ( (+)-catechin and epicatechin) levels were significantly higher in the spleens of experimental mice than in the controls. 4. Positive correlation between spleen cellularity and quercetin, and negative correlation between spleen cellularity and epicatechin content were observed. 5. Spleen mass and spleen lymphocytes phenotype and proliferation in RKW and RKW-A fed mice did not differ from the control. These results, together with suspicion of some embryo-toxicity, are worrying and eliminate the possibility of use Rhodiola kirilowii extracts for long-term treatment in pregnant females.

Vernonia polyanthes Less. (Asteraceae) is a medicinal plant that has been broadly used in Brazil for the treatment of inflammation and cutaneous damages. The present study evaluated the phytochemical contents and the antioxidant and topical anti-inflammatory activities of the V. polyanthes branches. Dried and powdered of V. polyanthes branches were exhaustively extracted with ethanol by static maceration followed by partition to obtain the hexane, dichloromethane, ethyl acetate, and butanol fractions. Phytochemical screening and the total phenolic and flavonoid contents were determined using chemical reactions and spectrophotometry, respectively. The antioxidant activity was evaluated by DPPH, reducing power of Fe +3 and β-carotene/linoleic acid assays. The topical antiinflammatory effects of the ethanol extract (EEVPB) and ethyl acetate fraction (EFVPB) were investigated in Croton oil-, arachidonic acid-, and phenol-induced ear edema models. Tannins, flavonoids, coumarins, terpenoids and steroids, saponins and alkaloids were detected in the ethanol extract and/or fractions. In these samples, the total phenolic and flavonoid contents varied from 0.65 ± 0.07 to 12.58 ± 0.67 g/100 g and 0.15 ± 0.02 to 4.75 ± 0.21 g/100 g, respectively. After topical application, EEVPB and EFVPB significantly (p < 0.01 or p < 0.001) reduced ear edema induced by three phlogistic agents (0.1, 0.5 or 1.0 mg/ear). These results suggest that V. polyanthes is an important and promising source of bioactive compounds with relevant biological properties and can be used as a strategy to develop new products for the treatment of skin conditions related to oxidative damage.
