Knockout of the BK β2 subunit abolishes inactivation of BK currents in mouse adrenal chromaffin cells and results in slow-wave burst activity

Abstract: Chromaffin cells from mice lacking the BK β2 subunit show decreased action potential firing during current injection but an increase in spontaneous burst firing.

“…One recent report showed that β1 deficiency exacerbated vascular fibrosis and remodeling (Xu et al, 2015). The knockout mice of β4 subunit showed signs of an epileptic phenotype (Brenner et al, 2005), and the β2 knockout mice had an increased tendency toward spontaneous burst firing in the adrenal medullary chromaffin cells (Martinez-Espinosa et al, 2014). A β3 (delA750) mutant, which led to truncation of the C-terminus, was reported to be associated with a form of generalized epilepsy (Lorenz et al, 2007).…”

“…As in many neurons, a fraction of CCs can undergo also spontaneous bursts at normal physiological conditions [22, 24]. Burst firing of APs occurs as a train of APs on top of a sustained plateau potential of variable duration (Fig.…”

“…As in many brain regions, BK channels are highly expressed in RCCs [10, 86], MCCs [9, 19, 22] and bovine CCs (BCCs) [85, 87], To activate during physiological depolarization, BK channels need an intracellular Ca 2+ concentration of at least 10 µM [84]. Such high concentrations occur only within “Ca 2+ -nanodomains” in the close vicinity of the Ca 2+ source [88].…”
