Krüppel-Like Factor 4 Acts as an Oncogene in Colon Cancer Stem Cell-Enriched Spheroid Cells

Abstract: Cancer stem cells (CSCs), a rare population in any type of cancers, including colon cancer, are tumorigenic. It has been thought that CSCs are responsible for cancer recurrence, metastasis, and drug resistance. Isolating CSCs in colon cancers is challenging, and thus the molecular mechanism regulating the self-renewing and differentiation of CSCs remains unknown. We cultured DLD-1 cells, one of types of cells derived from colon cancers, in serum-free medium to obtain spheroid cells. These cells possessed the c… Show more

“…Based on these results, KLF4 was suggested to function as a tumor suppressor. Accordingly, the overexpression of KLF4 was demonstrated to inhibit cell growth, migration, invasion and metastasis in liver, lung and colorectal cancer (29)(30)(31), and induce tumor cell apoptosis in esophageal cancer (32) and bladder cancer (33). These functional investigations also suggested that the loss of KLF4 expression may be involved in the initiation and formation of precancerous lesions in various cancer types.…”

“…Each of these factors has been linked to tumor aggressiveness and poor prognosis (Ben-Porath et al 2008;Kim et al 2013;Kareta et al 2015). MYC is one of the most thoroughly studied oncogenes (Cole 1986), and KLF4 has also been classified as an oncogene (Leng et al 2013). Recently, SOX2 was shown to maintain self-renewal and survival of CSCs in multiple tumor types, including squamous cell carcinoma (Boumahdi et al 2014).…”

“…Recently, Leng et al reported that Klf4 was over expressed in spheroid-cultured, cancer stem cell-like DLD-1 cells, possibly assigning Klf4 a tumor growth promoting role [43]. Possible effects on cancer stem cells were not investigated in our study, although we have previously shown that GLV-1h68 replicates preferentially in human breast cancer stem-like cells [44].…”
