KY1022, a small molecule destabilizing Ras via targeting the Wnt/β-catenin pathway, inhibits development of metastatic colorectal cancer

Abstract: APC (80-90%) and K-Ras (40-50%) mutations frequently occur in human colorectal cancer (CRC) and these mutations cooperatively accelerate tumorigenesis including metastasis. In addition, both β-catenin and Ras levels are highly increased in CRC, especially in metastatic CRC (mCRC). “…Bacterial cells were grown in LB media with ampicillin (100 lg/ml) at 37°C until the optical density at 600 nm reached approximately 0.6, at which time IPTG was added to a concentration of 0.2 or 1 mM, and the cells were incubated for an additional 20 h at 18°C. To prepare uniformly 15 N-and 13 C/ 15 N-labeled proteins for the NMR spectroscopy experiments, the cells were cultured in M9 media that contained 15 NH 4 Cl (Cambridge Isotope Lab) as the source of nitrogen and 13 C-Dglucose.
