Label‐Free Homogeneous Immunosensor Based on FRET for the Detection of Virus Antibody in Serum

Abstract: All lit up: A new label‐free immunosensor detects SV40 antibody (Ab). It is based on resonance energy transfer (FRET) from quantum dots (QDs) encapsulated in simian vacuolating virus 40 (SV40) virus‐like particles to graphene oxide. This method is rapid, inexpensive, and easy to perform.

“…Using a similar concept, a label-free homogeneous immunosensor for the detection of the simian vacuolating virus 40 (SV40) was developed. 802 QDs coated with SV40 virus-like particles (SVLP) were adsorbed to the surface of GO through hydrophobic interactions leading to ET and quenching of the QD PL. The addition of an anti-SV40 antibody created a larger complex with the SVLP-coated QDs that increased the distance between the QD and GO, reducing ET.…”

“…An original approach relies on an immunosensor made of GO and quantum dots encapsulated in simian vacuolating virus 40 (SV40) virus-like particles, which was exploited for the detection of anti-SV40 antibodies in serum. 202 SV40 is a nonenveloped tumorigenic virus, and the occurrence of nucleic acid sequences identical with SV40 sequences was observed in some tumors. As a result, the detection of serum anti-SV40 antibodies allows the diagnosis of cancer related to SV40.…”

“…The energy was transferred from SVLP-QD to GO, and the constructed label-free immunosensing platform could be used for rapid, inexpensive, and easy-to-perform detection of anti-SV40 antibody directly in serum. 156 Until now, several kinds of immunoassay systems including electrochemical and colorimetric ones were explored based on intrinsic properties of graphene, e.g., performing a central role in fast electron-transfer kinetics as well as promoting further signal amplication, for achieving high detection sensitivity, wide detection range, good selectivity and low detection limit. In the next step, real-life applications, such as cancer diagnostics, of these novel immunosensors prepared by graphene hybrids and composites integrated with both organic and inorganic materials should be carried out.…”
