Significance and Impact of the Study: This is the pioneer study on screening and isolation of antimicrobial peptide from the blood of Andrias davidianus. Here, we have developed a novel method by combining magnetic liposomes adsorption with reversed-phase high-performance liquid chromatography to purify and screen the antimicrobial peptides. From this screen, we identified a novel antimicrobial peptide which we name as andricin B. Andricin B is unique as it checks the growth of both Gram-positive and Gram-negative bacteria as well as few fungal species. AbstractThe Andrias davidianus has been known as a traditional Chinese medicine for a long time. Its blood is considered as a waste or by-product of the meat production industry. Although there are reports on isolation of the antimicrobial peptides from different resources, there are no reports of their isolation from A. davidianus blood. In this work, an antimicrobial peptide, andricin B, was isolated from the blood of A. davidianus by an innovative method in which the magnetic liposome adsorption was combined with reversed-phase high-performance liquid chromatography. The structure, antimicrobial activity and safety of andricin B were further investigated. Amino acid sequence was determined by N-terminal sequencing and found to be Gly-Leu-Thr-Arg-Leu-Phe-Ser-Val-Ile-Lys. Circular dichroism (CD) spectra and prediction of three-dimensional structure by bioinformatics software suggested the presence of a well-defined random coil conformation. Andricin B was found to be active against all bacteria tested in this study as well as some fungi. The minimum inhibitory concentrations (MICs) were in the range 8-64 lg ml À1 . Moreover, the haemolytic testing also suggested that andricin B could be considered safe at the MICs. Finally, andricin B was shown to inhibit the growth of Staphylococcus aureus in the cooked meat of A. davidianus. This study shows that andricin B is a promising novel antimicrobial peptide that may provide further insights towards the development of new drugs.

Yak butter is one of the most important foods for the Tibetan people. Of note, its production yields waste yak milk as a by-product. In this work, waste yak milk protein hydrolysates made via Pepsin hydrolysis were shown to have antimicrobial activity. Furthermore, an innovative method of magnetic liposome adsorption combined with reversed-phase high performance liquid chromatography (RP-HPLC) was developed to screen for and purify the antimicrobial peptides. Two antimicrobial peptides were obtained and their amino acid sequences were determined by N-sequencing, namely Arg-Val-Met-Phe-Lys-Trp-Ala and Lys-Val-Ile-Ser-Met-Ile. The antimicrobial activity spectra of Arg-Val-Met-Phe-Lys-Trp-Ala included Bacillus subtilis, Staphylcoccus aureus, Listeria innocua, Escherichia coli, Enterobacter cloacae and Salmonella paratyphi, while the Lys-Val-Ile-Ser-Met-Ile peptide shows not only bacterial growth inhibition but also of fungi. Haemolytic testing suggested that these two antimicrobial peptides could be considered to have no haemolytic effect at their minimum inhibitory concentrations (MICs).

Drug discovery and development programs have historically mainly focused on plants with medicinal properties and the extensive knowledge of traditional healers about these plants. More recently, the vast array of marine invertebrates and insects throughout the world have been recognized as additional sources for identifying unusual lead compounds to obtain structurally novel and mechanistically unique therapeutics. The results from these efforts are encouraging and have yielded a number of clinically useful drugs. However, many arthropods other than insects -such as spiders, scorpions, horseshoe crabs, sea spiders, centipedes, and millipedes -also produce hundreds of bioactive substances in their venom that may become useful in the clinic. Many of these chemicals are defensive and predatory weapons of these creatures and have been refined during millions of years of evolution to rapidly and with high specificity and high affinity shut down critical molecular targets in prey and predators. Exploration of these compounds may lead to the development of, among others, novel drugs for treating diseases caused by abnormalities in humans in the same (evolutionary conserved) molecular targets such as erectyle dysfunction, botulism, and autoimmune disorders, as well as the identification of novel antineoplastic, antimicrobial, and antiparasitic compounds. This paper addresses the importance of bioactive compounds from spiders, scorpions, horseshoe crabs, sea spiders, centipedes, and millipedes to these advances.
