Lactobacillus spp. and Enterococcus faecium strains isolation, identification, preservation and quantitative determinations from turkey gut content

Abstract: From gut content (ileum and cecum) of eight turkeys, 73 days old, was isolated, phenotypic identified and preserved, nine strains from Lactobacillus genus (L. acidophilus IBNA 11, L. acidophilus biotype 3 IBNA 12, L. fermentum biotype 1, IBNA 13-18 and L. salivarius IBNA 19) and one strain of Enterococcus faecium (IBNA 10).The parallel identification of strains was made by apiwebTM API50CHL V.5.1, BioMerieux (France) software, API20STREP, and ABIS online software. Quantitative level of Lactobacillus spp. strai… Show more

“…The intestinal content (ileum from 1 cm distal to Meckel's diverticulum to the ileo-cecal junction, and cecum) was aseptically removed and placed in sterile plastic bags on ice. Then, in the lab, 10-fold serial dilutions of 1 g of digesta (ileal and cecal content) were homogenized with 7 mL Brain Heart Infusion broth (Oxoid Ltd., England) supplemented with 2 mL glycerol, and immediately frozen at −20°C until the analysis, according to the technique previously described by Sorescu et al. (2019) .…”

“…The Mountzouris method (2007) completed by Sorescu et al, (2019) was applied. Sample preparation: 1 g intestinal content (ileum and cecum, respectively) per capita from seventeen chicks (Cobb 500, 45-day-old) was homogenized with 7 ml Oxoid BHI (Brain Heart Infusion) broth and 2 ml glycerol, and immediately frozen at -20 ºC until testing (no more than three months).…”

“…After defrost, decimal dilutions from every sample were inoculated on Oxoid Man, Rogosa, Sharpe (MRS) agar. Further instead, the procedure presented in other paper (Sorescu et al, 2019) for the isolation and counting of Lactobacillus CFU has been applied.…”

“…However, there are some molecular differences between mammalian IgG and bird IgY ( Table 1). The evolutionary distance between birds and mammals has made these two molecules have different properties [21,22]. For example, unlike IgG, IgY lacks a hinge region between the two "arms" of the antibody (Figure 1).…”
