Lactoferrin inhibits the inflammatory and angiogenic activation of bovine aortic endothelial cells

Abstract: Lf exerted a potent effect on BAEC activation, suggesting that it might function via an endothelia-based mechanism in the treatment of various diseases, including rheumatoid arthritis and cancer.

“…5 Furthermore, bLF has been shown to suppress angiogenic activation in bovine aortic ECs. 7 Thus, the effects of LF on angiogenesis remain controversial, although the results of this study indicate that bLF activates endothelial function and promotes an ischemia-induced angiogenic response. Although it is difficult to explain why our results are in opposition to those of previous studies, 1 plausible reason is that the doses of bLF used were different, ranging from 0.01 to 1 mg/mL in our study versus 10 to .100 mg/mL in others.…”

“…Cells were then transferred to 96 well plates coated with Matrigel Basement Membrane Matrix (LDEV-free, Corning) in triplicate and cultured in EBM media (Lonza). Formation of tubes by human microvascular endothelial cells was examined 18hrs later using HMVECs cultured in Matrigel as described [23]. The tube number and tube length per well were counted using NIS Elements software (Nikon).…”

“…IL-1B increases the activity of hypoxia-inducible factor (HIF)-1, which promotes the production and release of VEGF. 16,17 It is thought that the inhibition of these cytokines occurs via LF interference with nuclear transcription factor B (NF-kB). 18 Oral administration of bovine LF in mice increases levels of IL-18, an anti-angiogenic cytokine with possible direct action on endothelial cells.…”
