Large Volume Electron Microscopy and Neural Microcircuit Analysis

Abstract: One recent technical innovation in neuroscience is microcircuit analysis using three-dimensional reconstructions of neural elements with a large volume Electron microscopy (EM) data set. Large-scale data sets are acquired with newly-developed electron microscope systems such as automated tape-collecting ultramicrotomy (ATUM) with scanning EM (SEM), serial block-face EM (SBEM) and focused ion beam-SEM (FIB-SEM). Currently, projects are also underway to develop computer applications for the registration and segm… Show more

“…Classical EM techniques had clear limitations with respect to efficiency and the use of fixation and staining methods that might not always represent axonal structure in all accuracy. New techniques such as large-volume EM or cryo-electron tomography ( Kubota et al, 2018 ; Xu et al, 2017 ; Chakraborty et al, 2020 ) provide ever-improving means to drive the discovery processes toward good understanding of axon structure, which will eventually have important implications for work on neuronal pathologies.…”

“…For example, a 2-week reduction in physical activity levels causes a decrease in cardiorespiratory fitness (Bowden Davies, 2018) and a week of sedentary behavior, which leads to impairment in mood and depression (Edwards and Loprinzi, 2016). Also, sedentary behavior, for example, screen time, is a vital risk factor for venous thromboembolism (Kubota et al, 2018). However, the regular practice of physical activity and decreased levels of sedentarism are associated with reduced risk for morbidity and mortality (Ekelund et al, 2019).…”

“…Over the past decade, technological improvements such as improved direct electron detectors have enabled the practical applications of techniques such as volume electron microscopy [2][3][4][5][6]. Advances in microscopy techniques in recent years have opened windows into cells, giving us insight into cellular organization with unprecedented detail [7][8][9][10][11][12][13]. Volumetric EM enables the capture of 3D ultrastructural datasets (i.e., images where fine structures such as membranes of cells and their internal organelles are resolved, as shown in Fig 1A).…”
