Laser confers less embryo exposure than acid tyrode for embryo biopsy in preimplantation genetic diagnosis cycles: a randomized study

Abstract: We compared two methods of zona pellucida drilling. 213 embryos were biopsied with acid Tyrode. Each biopsy took 3 minutes and the entire procedure ~29 minutes. 5% of blastomeres lysed, 49% of embryos became blastocyst and 36% of patients became pregnant. 229 embryos were biopsied with laser. Each biopsy took 30 seconds and the entire procedure ~7 minutes. 2.5% of blastomeres lysed, 50.6% of embryos became blastocyst and 47% of patients became pregnant. We can conclude that laser can be used for embryo biopsy.… Show more

“…It can be achieved through laser pulses [19], Tyrode acid [20] or mechanically [21]. The clinical results after the use of these three methods are comparable, as it has been reported in several RCTs on sibling embryos [22][23][24]. However, laser-based zona drilling is the most standardized and reproducible approach.…”

“…However, laser-based zona drilling is the most standardized and reproducible approach. Indeed, it is less operator-dependent [24] and less time-consuming and a shorter learning curve is required to acquire this skill. Nevertheless, the considerable raise in the temperature of the culture media surrounding the embryo during the firing procedure [25] was perceived as a concern.…”

“…Terlebih dahulu embrio dipastikan tidak dapat bergerak dengan menahannya pada holding pipet. Pada saat yang hampir bersamaan pipet kedua dengan diameter 10 mikro yang beirisi larutan asam tyrode didekatkan pada zona pelusida dan dibuat lubang dengan menyemprotkan secara terkontrol larutan asam tyrode pada permukaan zona (Geber et al 2011).…”

“…Various techniques (i.e., mechanical dissection, chemical drilling, and laser ablation) for opening of the zona pellucida (ZP) have been developed [ 4 - 8 ]. However, not all of them have the same potential of success for PGD because the viability of embryo and/or blastomere may be affected by the biopsy procedure [ 9 - 12 ].…”
