Lateral hypothalamic NMDA receptor subunits NR2A and/or NR2B mediate eating: immunochemical/behavioral evidence

Abstract: Cells within the lateral hypothalamic area (LHA) are important in eating control. Glutamate or its analogs, kainic acid (KA) and N-methyl-d-aspartate (NMDA), elicit intense eating when microinjected there, and, conversely, LHA-administered NMDA receptor antagonists suppress deprivation- and NMDA-elicited eating. The subunit composition of LHA NMDA receptors (NMDA-Rs) mediating feeding, however, has not yet been determined. Identifying this is important, because distinct second messengers/modulators may be acti… Show more

“…In contrast, the BSTdm projections to the supraoptic nucleus are found caudally and ventrally, where vasopressinergic neuronal cell bodies predominate-as well as rostrally and dorsally where oxytocinergic cell bodies predominate. The BSTdm also densely innervates several clusters of magnocellular neurons forming part of the accessory supraoptic nucleus, where complex mixtures of oxytocinergic and vasopressinergic neurons may be found (Peterson, 1966;Duan and Ju, 1998;Khan et al, 1999).…”

“…anesthesia, each rat was stereotaxically fitted with a unilateral, stainless-steel indwelling guide cannula [26 gauge; outer diameter (o.d. ), 0.46 mm] targeted toward the PVH, using the general stereotaxic procedure described by Khan et al (1999). With the incisor bar 3.3 mm below the ear bars, stereotaxic coordinates (Paxinos and Watson, 1998) were as follows: 6.8 mm anterior to the interaural line, 0.4 mm lateral to the midsagittal sinus, and either 6.4 or 7.2 mm ventral to the surface of the skull.…”

“…The effect of glutamate antagonists on feeding has been variable, depending on the site tested and the type of glutamate receptor blocked (72)(73)(74). Currently, there are insufficient data about the effects of these drugs on the hypothalamus or other areas of the brain that mediate feeding and hunger to know how these drugs reduce weight.…”
