Lectin binding characteristics of mouse placental cells

Abstract: The lectin binding characteristics of mouse placental cells were examined. Wax embedded tissue sections of placentae from d 14 pregnant mice were stained with 26 lectins, with a wide range of sugar specificities. Cell cultures prepared from d 14 mouse placentae and cultured for 24 h were stained with 7 of the lectins to determine if they could be used as markers for the different trophoblast cells in culture. In tissue sections all placental cell populations bound lectin but no lectin bound specifically to any… Show more

“…Arachis hypogaea what was available in the bibliography (Bulmer & Peel, 1996;Díaz et al, 2017;Dorsch et al, 2019;Fernández et al, 2000Fernández et al, , 2014Fiorentino et al, 2018;Jones et al, 1995Jones et al, , 1997Jones et al, , 2007Jones et al, , 2014Jones et al, , 2016Jones & Aplin, 2009;Munson et al, 1989;Sandoval et al, 2001;Stewart et al, 2000;Sgambati et al, 2002;Tatsuzuki et al, 2009) and from our observations in L. maximus. We used a tree based on the taxon-rich and well-resolved phylogeny of Placentalia (O'Leary et al, 2013) combined with a more comprehensively sampled phylogeny of Rodentia (Fabre et al, 2015).…”

“…A relevant feature related to the invasiveness of the trophoblast and its expulsion at birth is the glycotype, a specific expression of different glycosidic residues (Jones et al, 1997). The glycotype of placentas with different invasiveness degrees (Jones et al, 1995, 2000, 2002, 2004, 2007, 2009, 2014; Stewart et al, 2000) has been determined using lectin histochemistry. Its knowledge allows the understanding of variations in unusual pregnancy events, such as the physiological early embryonic death (Fiorentino et al, 2018; Díaz et al, 2017; Dorsch et al, 2019), and the phylogenetic relationships between clades (Jones et al, 2007; Klisch et al, 2010).…”

“…Complementary characterization of the morphology of the junctional zone was performed using immunohistochemistry techniques for isolectin B4 (Figure 7, lower panels). Isolectin B4 has greater specificity for labyrinth-derived cells and leaves the junctional zone primarily devoid of stain [40]. Placental tissues derived from Ubp43 null fetuses had large infarcted areas in contrast to wt Ubp43 fetal-derived placentas.…”

“…Glycoconjugate contents in cytoplasmic granules of uNK cells have been demonstrated to react with Shiff's reagent following periodate treatment, therefore, Periodic acid-Schiff (PAS) staining was established as a method of uNK-cell identification in the mouse and human endometrium ( 11 , 12 , 15 ). However, the more recently developed dolichos biflorus agglutinin (DBA) lectin staining was demonstrated to be a more sensitive method for the identification and localization of uNK cells ( 16 , 17 ). DBA lectin can react with glycoconjugates within the granules and at the surface of uNK cells.…”
