Lessons from a tarantula: new insights into myosin interacting-heads motif evolution and its implications on disease

Abstract: Tarantula's leg muscle thick filament is the ideal model for the study of the structure and function of skeletal muscle thick filaments. Its analysis has given rise to a series of structural and functional studies, leading, among other things, to the discovery of the myosin interacting-heads motif (IHM). Further electron microscopy (EM) studies have shown the presence of IHM in frozen-hydrated and negatively stained thick filaments of striated, cardiac, and smooth muscle of bilaterians, most showing the IHM pa… Show more

“…Thus, BS binding is compatible with the sequestered state. The fact that BS favors the off state 40 is a strong argument in favor of the model previously proposed 41 from low resolution EM maps which we have defined at a better resolution here with both heads mainly unaffected for the motor domain but which uses hinges at the converter/ELC interface to allow the asymmetric association of the myosin heads to form the IHM (Fig. 6).…”

“…A second cautionary note is that while purified smooth muscle HMM has been shown to form an IHM structure by high-resolution electron microscopy [ 96 , 97 ], most studies related to the IHM structure are based on low-resolution (≥ 2 nm) EM reconstructions from isolated myosin thick filaments (for reviews, see [ 6 , 88 ]). It is imperative that the high-resolution structure of the folded-back sequestered state of human β-cardiac myosin be determined.…”

“…This single molecular entity found in tarantula thick filaments was called the myosin interactingheads motif (IHM) (Alamo et al 2008) and its consequences on structure and function are the focus of this review. We review the IHM evolution and its implications on disease in Alamo et al (2017a).…”
