Lessons from the Embryonic Neural Stem Cell Niche for Neural Lineage Differentiation of Pluripotent Stem Cells

Abstract: Pluripotent stem cells offer an abundant and malleable source for the generation of differentiated cells for transplantation as well as for in vitro screens. Patterning and differentiation protocols have been developed to generate neural progeny from human embryonic or induced pluripotent stem cells. However, continued refinement is required to enhance efficiency and to prevent the generation of unwanted cell types. We summarize and interpret insights gained from studies of embryonic neuroepithelium. A multitu… Show more

“…Differentiating hPSCs into OPCs or neural progenitor cells (NPCs) with a high purity has been demonstrated, but their function and maturation are still under investigation [12][13][14] . Extracellular matrix (ECM) plays an important role in neural differentiation of hPSCs and the maturation of the derived neural cells [15] . ECM proteins, through the interaction with integrins expressed on the neural cells, modulate cell survival, migration, proliferation, and the differentiated cell function [16] .…”

“…A recent publication has exploited the midbrain floorplate precursor marker CORIN 34,35 to enrich for dopaminergic precursors in Parkinson cell transplantation paradigms 36 . CD molecules are not only markers, but functionally relevant mediators of cell-cell interactions and of a cell's ability to respond to cues from extracellular matrix molecules and growth factors 37 . One strategy of further enhancing the arsenal of combinatorial CD antigens to characterize neural lineage development is to use known intracellular markers to screen for and define CD antigen combinations for a particular cell type of interest.…”
