LGP2 Expression is Enhanced by Interferon Regulatory Factor 3 in Olive Flounder, Paralichthys olivaceus

Abstract: In innate immunity, LGP2 (laboratory of genetics and physiology 2) plays a very important role in the production of type I interferon (IFN) through recognition of cytosolic viral RNA. Although viral infection or stimulation with double-strand RNA dramatically induces expression of the LGP2 gene, the underlying transcriptional mechanism has never been studied. Here, we cloned and characterized the 5′-upstream region (−1,337 bp) of the LGP2 gene in olive flounder (Paralichthys olivaceus). Numerous canonical moti… Show more

“…olivaceus , C . idella and O. mykiss , expression levels of the lgp2 gene and type I ifn gene were dramatically induced by poly(I:C) stimulation and viral infection (Ohtani et al , ; Huang et al , ; Chang et al , ; Hikima et al , ). There is conflicting evidence, however, for example, in C .…”

“…In bony fishes such as P. olivaceus, C. idella and O. mykiss, expression levels of the lgp2 gene and type I ifn gene were dramatically induced by poly(I:C) stimulation and viral infection (Ohtani et al, 2010;Huang et al, 2010;Chang et al, 2011;Hikima et al, 2012). There is conflicting evidence, however, for example, in C. auratus that lgp2 inhibits poly(I:C)-induced activation of D. rerio type I ifn production (Sun et al, 2011).…”

“…DEGs from both challenge groups revealed 46 up-regulated genes expressed in both treatments (Fig.2C, Table S2, Data S1). Interestingly, cod, flounder (Paralichthys olivaceus) (22), chicken (Gallus gallus) (23) and the tree shrew (Tupaia chinensis) ( Table S3). Synteny in the teleost lineage was less conserved for ifih1-adjacent genes including fap, gcg and dpp4, which displayed significant variation.…”

“…2). Japanese flounder IRF3 regulates the transcription of type I IFN and laboratory of genetics and physiology 2 (LGP2) after viral infection or poly (I:C) treatment [37,38]. Hu et al [35] demonstrated that IRF3 expression in Japanese flounder reared at 18 C and treated with poly (I:C) could be detected at 1e4 days post injection (dpi) and then declined at 5e7 dpi.…”
