Life Style-Related Diseases of the Digestive System: Endocrine Disruptors Stimulate Lipid Accumulation in Target Cells Related to Metabolic Syndrome

Abstract: Abstract. Many reports indicated that endocrine disruptors (EDs) affect several hormonal functions in various living things. Here, we show the effect of EDs on lipid accumulation in target cells involved in the onset of metabolic syndrome. Treatment with nonylphenol and bisphenol A, typical EDs, stimulated the accumulation of triacylglycerol in differentiated adipocytes from 3T3-L1, preadipocytes, in time-and concentration-dependent manners. Up-regulation of gene expressions involved in lipid metabolism and me… Show more

“…Obesity occurs by adipocyte hypertrophy or hyperplasia. Hypertrophy seems to be the initial steps towards adipocyte cell dysfunction (Monteiro, 2009;Monteiro et al, 2006) and POPs action on these mechanisms may promote their dysregulation (Grun and Blumberg, 2009;Wada et al, 2007). Our data did not reveal a correlation between the ΣPOP levels and adipocyte size from the corresponding tissue, probably because we analysed an obese population, and adipocytes are at the limit of lipid accumulation (Tan and Vidal-Puig, 2008;Virtue and Vidal-Puig, 2010).…”

“…There is a complex relationship between prenatal exposure to oestrogen and offspring weight at maturity [70]. BPA has been found to stimulate lipid accumulation and the differentiation of preadipocytes into mature cells [71]. It seems that PPAR may be a link to explain the molecular mechanisms of EDC impact on the pathogenesis of obesity.…”

“…Istnieje zależność pomiędzy prenatalną ekspozycją na estrogeny a masą ciała potomstwa w okresie dojrzałości [70]. Wykazano, że BPA również wykazuje zdolność do stymulacji odkładania lipidów w adipocytach oraz wpływa na dojrzewanie preadipcytów [71]. Wydaje się, że molekularny mechanizm działania EDC poprzez receptory PPAR może tłumaczyć ich negatywną rolę w patogenezie otyłości.…”

“…Both sex steroids and leptin are involved in early organizational effects in the hypothalamus that affect respectively the central control of reproduction [40 • ] and energy balance [41 • ]. Peripherally, whereas EDC effects on male and female reproductive tract have been extensively described [2 • ], EDCs appeared more recently to cause induction of cell differentiation and adipogenesis [42] as well as lipid accumulation in differentiated adipocytes [43]. Gestational and lactational exposure to BPA resulted in increased adipogenesis at weaning in female rats [44].…”
