Linking Immunity, Epigenetics, and Cancer in Inflammatory Bowel Disease

Abstract: Most of what is known about the pathogenesis of inflammatory bowel disease (IBD) pertains to complex interplay between host genetics, immunity, and environmental factors. Epigenetic modifications play pivotal roles in intestinal immunity and mucosal homeostasis as well as mediating gene-environment interactions. In this article, we provide a historical account of epigenetic research either directly related or pertinent to the pathogenesis and management of IBD. We further collate emerging evidence supporting r… Show more

“…The proinflammatory cytokine Interluekin-1β (IL-1β), secreted from immune cells within the lamina propria, also stimulates DNMT1 activity in a nitric oxide (NO)-dependent manner through upregulation of inducible nitric oxide synthase (iNOS) in IECs ( Fig. 1) 23,46) . “…Alterations to chromatin are central to the reprogramming of pathological gene expression, which is directly relevant to many inflammatory diseases, including IBD (2,13,14). Studies have shown that DNA methylation is well correlated with various aspects of IBD, including disease duration and severity of inflammation (15,16).…”

“…Inflammatory bowel disease has complex pathogenesis involving genetic susceptibility, intestinal microbiota, the host immune system and environmental factors such as diet, stress, smoking and hygiene (26). Defects in signalling pathways can lead to dysregulation of the inflammatory response that are crucial in the pathogenesis of IBD.…”
