Linking Spermatid Ribonucleic Acid (RNA) Binding Protein and Retrogene Diversity to Reproductive Success

Abstract: Spermiogenesis is a postmeiotic process that drives development of round spermatids into fully elongated spermatozoa. Spermatid elongation is largely controlled posttranscriptionally after global silencing of mRNA synthesis from the haploid genome. Here, rats that differentially express EGFP from a lentiviral transgene during early and late steps of spermiogenesis were used to flow sort fractions of round and elongating spermatids. Mass-spectral analysis of 2D gel protein spots enriched >3-fold in each fractio… Show more

“…Spermatogenic stages were scored in paraformaldehyde-fixed, frozen histological sections (section thickness, 8 lm) prepared from testes of wildtype rats after staining by the periodic acid-fuchsin sulfurous acid technique [28] but modified for frozen sections (John M. Shelton and James A. Richardson, UTSW Molecular Pathology Core Laboratory) [29]. Spermatid development was used as the primary criterion to identify cellular associations defining each of the 14 spermatogenic stages in Sprague-Dawley rats [5,7,30].…”

“…PSIP1, another RNA binding protein, was expressed in spermatocytes and spermatids. It has been reported that PSIP1 is closely involved in spermatomiosis [ 31 ]. Sam68 has also been found to be closely related to spermatogenesis and plays an important role in regulating the expression of meiosis-related genes [ 32 , 33 ].…”

“…PCBP3 (BTA1 at 147 Mb), KCNK10 (BTA10 at 100 Mb), and MRPS5 (BTA11 at 1 Mb) emerged as candidate genes for PPAI. PCBP3 protein was expressed in spermatids of rats during spermiogenesis (Chapman et al, 2013). KCNK10 was associated with semen volume in bulls (Hering et al, 2014) and was expressed at the membrane of oocytes Table 1.…”
