LIPID COMPOSITION OF QUAKING MOUSE MYELIN: COMPARISON WITH NORMAL MOUSE MYELIN IN THE ADULT AND DURING DEVELOPMENT1

Abstract: Abstract— In the ‘Quaking’ mouse, a deficiency in the long chain fatty acid content of galactolipids has been shown to occur. Myelin in the mutant has been compared to myelin in adult and in 12‐day‐old controls. We have shown that myelin is not only quantitatively reduced but also qualitatively modified, with a higher protein and a lower galactolipid content. Cerebrosides contain only a small amount of kerasin, lacking long chain nonhydroxylated fatty acids in comparison to both controls; the relative percenta… Show more

“…The curve here presented shows that the synthesis of stearic acid by the first elongating complex (Ci6-elongase) occurs somewhat before the biosynthesis of very long chain (Ci8-elongase). This is in agreement with the composition of young my elin which is richer in medium chain than the adult one (19). As previously shown (20), the cytosolic de novo system is optimum at 6 days after birth.…”

“…Forebrain myelin was isolated (10) and its purity controlled (11). Lipids and fatty acids were extracted, identified and measured as previously described (12).…”

“…As it is easy to isolate pure myelin from frozen brains (O'Brien and Sampson, 1965;Everly et al. 1977;Bourre et al, 1978b), electron microscopy was performed as previ ously described (Baumann et at., 1973). Subfractions of myelin were obtained by successive sucrose gradi ents of varying molarity (Bourre et al, 1977).…”
