Liquid extraction surface analysis nanospray electrospray ionization based lipidomics for in situ analysis of tumor cells with multidrug resistance

Abstract: Lower cell membrane fluidity and higher cell membrane potential caused by the changes in lipid compositions, enhanced anti-oxidative ability and energy generation were involved in the development of the MDR. The specific alterations identified in this study may provide more information for overcoming MDR.

“…Another group identified 24 biomarkers and found the pathways of protein and cysteine synthesis, glutamine metabolism and the ammonia cycle were related to drug resistance by using MCF-7/ADR and its parental MCF-7/S cells with ultra-performance liquid chromatography-tandem mass spectrometry 75. Moreover, the ratios of GSH/GSSG, ATP/ADP and ATP/AMP were prominently higher in MCF-7/ADR cells compared to MCF-7/S cells 76. Statins are considered to have antitumour potential in recent years 77.…”

“…179 Zong et al have employed LESA-MS for the in situ analysis of lipids in breast cancer cells dried on glass microscope slides, permitting the characterization of multidrug resistance based on changes in cellular lipid profiles. 180 The integration of atomic force microscopy for imaging cell morphology prior to and postanalysis indicated structural differences of cells post-LESA sampling, revealing the effective extraction of lipids from the cells. Another study profiled lipids in mycobacterial cells directly from colonies on agar media.…”

“…The various liquid extraction techniques include liquid extraction surface analysis (LESA) [8], nanospray desorption electrospray ionisation (nano-DESI) and devices such as the FlowProbe (Prosolia Inc. Indianapolis, IN) and the MasSpec Pen [9]. In the last decade, these techniques have become established for analysis of small molecules, metabolites and lipids directly from biological samples (LESA [10][11][12], nano-DESI [13][14][15], FlowProbe [16], MasSpec Pen [17]), but in situ protein analysis has proven a greater challenge.…”
