Liver‐Target and Glucose‐Responsive Polymersomes toward Mimicking Endogenous Insulin Secretion with Improved Hepatic Glucose Utilization

Abstract: Oral insulin therapy that targets the liver and further mimics glucoseresponsive secretion holds promise for correcting defects in glucose metabolism caused by peripheral delivery. This work describes the construction of polymersomes (Pep-PMS), which are composed of glucose-responsive polymers decorated with peptides that readily bind to the ganglioside-monosialic acid (GM1) receptor in the intestinal epithelium. Pep-PMS are efficiently transported across the intestinal epithelium through GM1-mediated transcyt… Show more

“…S5). The UiO-68-NH 2 NPs exhibited a high LC of 33 weight % (wt %) at the feeding ratio of 4:1, higher than most of the INS nanocarriers being reported (table S1) (29,51). The mass loss was also observed to determine the LC of the UiO-68-NH 2 -based nanosystem by thermogravimetric analysis (TGA), which was consistent with the above results (fig.…”

“…These nanosystems were able to release the encapsulated insulin in hyperglycemic conditions based on GOx-induced H 2 O 2. These orally-delivered, liver targeting systems were successful in regulating insulin delivery in rat models, and could be translated favorably into clinical praxis [ 179 ]. Lectin is a group of naturally glucose- and mannose-binding protein moieties.…”
