Local actin polymerization during endocytic carrier formation

Abstract: Extracellular macromolecules, pathogens and cell surface proteins rely on endocytosis to enter cells. Key steps of endocytic carrier formation are cargo molecule selection, plasma membrane folding and detachment from the cell surface. While dedicated proteins mediate each step, the actin cytoskeleton contributes to all. However, its role can be indirect to the actual molecular events driving endocytosis. Here, we review our understanding of the molecular steps mediating local actin polymerization during the fo… Show more

“…Various mechanisms of PEV formation and their scission from the cytoplasmic membrane have been suggested (Johannes et al 2016;Hinze and Boucrot 2018;Renard et al 2018;Sandvig et al 2018). PEVs usually have a round or a rounded elongated shape with a diameter from tens of nanometers to a few micrometers; the membrane of large vesicle can be plicated (with secondary invaginations) (Doherty and McMahon 2009;Hinze and Boucrot 2018). Endocytic plasmalemma invagination can also be as a long narrow tube (PET -primary endocytic tube; synonym for PEV and PET -endocytic carrier).…”

“…Endocytic plasmalemma invagination can also be as a long narrow tube (PET -primary endocytic tube; synonym for PEV and PET -endocytic carrier). In some cases, PET is a stable formation inherent in a certain type of endocytosis (FEME, for example) (Hinze and Boucrot 2018); but more often, PET results from vesicle scission delay from the plasmalemma with active elongation of the neck of the membrane invagination for some causes. In experimental studies, the formation of PETs is artificially stimulated by various methods (by actin depolymerization and hyperosmotic shock, overexpression of BAR proteins, suppression of dynamin, exposition in GTP-γS, etc.)…”
