Long-Lasting Inhibitory Effects of Distigmine on Recombinant Human Acetylcholinesterase Activity

Abstract: To elucidate the mechanism whereby distigmine, an underactive bladder remedy, potentiates urinary bladder contractions long-lastingly, the inhibition of recombinant human acetylcholinesterase (rhAChE) by distigmine was investigated. “…For experiments, the thawed rhAChE stock solution was diluted 200-fold with modified Hanks' balanced salt solution of the following composition (in mmol/L) NaCl, 136.9; KCl, 5.37; CaCl 2 , 1.26; MgCl 2 , 0.81; Na 2 HPO 4 , 0.77; KH 2 PO 4 , 0.44; NaHCO 3 , 4.17; glucose, 5.55. rhAChE aliquoted to 96-well plates (0.025 μg/well) decomposed 0.952 mmol/L acetylthiocholine (ATCh) to produce thiocholine (TCh) at a rate of 1.28 ± 0.05 µmol/L/min, which was expressed as TCh concentration according to the obtained calibration curve. rhAChE activity was measured as previously described [10]. The diluted rhAChE solution (50 μL) and 5 mmol/L 5,5′-dithio-bis-(2-nitrobenzoic acid; DTNB, 149 μL) were added to each well of a 96-well plate and incubated for 10 min at 30 ° C. Subsequently, the test antipsychotics and neostigmine (2 × 10 -3 mol/L, 1 μL) were added to their corresponding wells, so that their final concentrations were 10 -5 mol/L and incubated for 30 min at 30 ° C.…”
