Backgroud: To elucidate the relationship between the expression level of HOXA-AS2 and it’s prognostic value of cancer by meta-analysis. Methods: Databases of PubMed, Cochrane Library, Embase, Web of Science, Google Scholar, CNKI and some others were searched systematically. Comprehensively screen according to the inclusion criteria and the exclusion criteria was conducted. The connection between HOXA-AS2 and the prognosis characteristics of patients with various types of cancer was screened by combining odds ratio (OR), Hazard ratios (HR) and 95% confidence interval (CI) for the studies collected in this meta-analysis. In addition, we further analyzed the expression of the gene and its potential clinical value in Gene Expression Profiling Interactive Analysis (GEPIA) and the lnCAR database. Results: A total of 12 studies consisting 796 patients were included in this research. Compared low HOXA-AS2 expression group, patients with high HOXA-AS2 expression were more likely to get poor overall survival (OS). Moreover, high HOXA-AS2 expression demonstrates advanced TNM stage, earlier lymph node metastasis, distant metastasis and bigger tumor size. But there was no correlation or the correlation was not statistically significant between the expression and the age, sex or the pathological differentiation. In addition, data from the GEPIA and LnCAR databases revealed that increasing HOXA-AS2 expression means bad prognosis in most of cancers. Conclusions: High HOXA-AS2 expression shows worse cancer prognosis in cancer patients, and HOXA-AS2 may be acted as therapeutic target and promising prognostic marker.

Background: Cervical cancer, the leading cause of cancer-relevant mortality in females, is an aggressive malignant tumor. Tumor angiogenesis is vital for cell proliferation and metastasis in cancers. Accumulating studies have claimed that long non-coding RNAs (lncRNAs) participate in the progression of various cancers. The aim of this research is to explore the biological role and regulatory mechanism of LncRNA HOXA-AS2 in cervical cancer.Methods: Experiments including RT-qPCR, western blot, RIP, MTT, EdU, transwell, luciferase reporter, RIP, FISH, tube formation assays were applied to investigate the biological role and regulatory mechanism of LncRNA HOXA-AS2 in cervical cancer.Results: In current study, the results disclosed that HOXA-AS2 was notably upregulated in cervical cancer tissues and cell lines, and high HOXA-AS2 expression was strongly associated with poor prognosis of cervical cancer patients. Furthermore, HOXA-AS2 contributed to cell proliferation, migration, invasion and angiogenesis in cervical cancer. In addition, HOXA-AS2 absorbed miR-509-3p and miR-509-3p targeted BTN3A1 in cervical cancer. Besides, BTN3A1 overexpression partly rescued the inhibitory influence of HOXA-AS2 knockdown on cervical cancer progression and angiogenesis. Overall, HOXA-AS2 promoted cervical cancer progression and angiogenesis through sponging miR-509-3p to elevate BTN3A1 expression. Conclusions: In other words, this paper was the first to study the molecular regulatory mechanism of HOXA-AS2 in cervical cancer and certified that HOXA-AS2 accelerated progression and angiogenesis in cervical cancer by targeting miR-509-3p/BTN3A1 axis, which may become a beneficial therapeutic target for cervical cancer.

Background: HOXA-AS2, a lncRNA, had been proved to exert the promoting effect on glioblastoma, but its regulatory mechanism was not fully revealed. Our study focused on the interaction and role of HOXA-AS2/miR-885-5p/RBBP4 axis in the development of glioblastoma. Methods: qRT-PCR was used to detect the expression of lncRNA, miRNA and mRNA in glioblastoma tissues and cells. Dual luciferase assay, RIP assay and RNA pull-down assay were applied to reveal the interaction among HOXA-AS2, miR-885-5p and RBBP4. The cell functional experiments such as CCK-8, BrdU assay and cell adhesion etc. were used to analyze the effect of HOXA-AS2/miR-885-5p/RBBP4 axis on glioblastoma cells. Results: HOXA-AS2 and RBBP4 were overexpressed in 33 glioblastoma tissue samples and glioblastoma cells, as well as HOXA-AS2 and RBBP4 contributed to the tumorigenesis of glioblastoma cells. On the other hand, miR-885-5p was proved to be down-regulated in 33 glioblastoma tissue samples and glioblastoma cells, and it was an obstacle to the malignant phenotypes of glioblastoma cells. Meanwhile, HOXA-AS2 could negative regulate miR-885-5p, thereby promoting the RBBP4 expression since RBBP4 was targeted by miR-885-5p. Finally, HOXA-AS2 was proved to act as the positive role in glioblastoma cells via negative regulating miR-885-5p to release RBBP4.Conclusion: HOXA-AS2 promotes the tumorigenesis of glioblastoma through targeting miR-885-5p to induce the RBBP4 expression.
