Long Noncoding RNA MIR100HG Knockdown Attenuates Hepatocellular Carcinoma Progression by Regulating MicroRNA-146b-5p/Chromobox 6

Abstract: Purpose. Hepatocellular carcinoma (HCC) accounts for approximately ninety percent of primary liver cancer. This study attempted to investigate the effects of the long noncoding RNA MIR100HG (MIR100HG) in HCC and the underlying molecular mechanism. Materials and Methods. qRT-PCR was implemented to analyze the expression of MIR100HG, microRNA-146b-5p (miR-146b-5p), and Chromobox 6 (CBX6). The correlation between MIR100HG and clinicopathological features of HCC patients was assessed. Additionally, the effects of … Show more

“…LncRNA MIR100HG was dysregulated in many cancers and played distinctively complex and contradictory roles, which involved tumorigenesis, proliferation, and invasion or inhibition of these biological behaviors of tumors (Wu et al, 2022). Previous studies had demonstrated that lncRNA MIR100HG was identified as an oncogene in various cancers, including breast cancer (Chen et al, 2020), hepatocellular carcinoma (HCC) (Li et al, 2021), gastric Frontiers in Genetics frontiersin.org cancer (Li J. et al, 2019;Li et al, 2020), and colorectal cancer (Li W. et al, 2019). However, studies in MIR100HG also proved that it could exert tumor suppressive effects in papillary-thyroid carcinoma (Yang et al, 2021;Park and Lee, 2022) and cervical cancer (Lang et al, 2022;Lin et al, 2022).…”

“…MIR100HG plays an important role in human tumors and is an lncRNA that has received a lot of attention in the tumors, and it is expressed differently in tumor tissues dependent on the type of tumor. MIR100HG is involved in tumor proliferation, migration, and invasion in breast cancer [ 34 ], liver cancer [ 35 ], and laryngeal squamous cell carcinoma [ 36 ]. Down-regulation of MIR100HG expression in gastric cancer inhibits proliferation, migration, and invasion of gastric cancer cells [ 37 ].…”

“…Subsequently, it is predicted that miR-146b-5p regulates both SLFN5 and MEG3 by RNA hybridization and confirmed that MEG3 can directly bind to miR-146b-5p by RNA pull down assay. However, miR-146b-5p showed the dual functions of promoting or inhibiting tumorigenesis in cancer [41][42][43][44][45]. For instance, Qu L et al reported that miR-146b-5p can promote Ewing's sarcoma malignancy by inhibiting B-cell translocation gene 2 expression [45].…”
