LncRNA PITPNA-AS1 was a newly identified lncRNA which has never been studied in cancers. Whether PITPNA-AS1 participated in the development of hepatocellular carcinoma (HCC) is obscure. Given the coaction of lncRNAs and miRNAs to carcinogenesis, the purpose of the present research is to inquire how PITPNA-AS1 affects HCC progression. Firstly, PITPNA-AS1 was observed to be heightened in HCC tissues. Then function assays proved that overexpressing or silencing PITPNA-AS1 could manipulate the proliferation and motility of HCC cells. Besides, PITPNA-AS1 was located in the cytoplasm. Among the candidate miRNAs of PITPNA-AS1, miR-876-5p was an obvious target. Moreover, mechanism experiments validated that PITPNA-AS1 modulated WNT5A expression by targeting miR-876-5p. Rescue experiments affirmed that WNT5A silencing rescued the miR-876-5p suppression-induced cellular processes in PITPNA-AS1-silenced Hep3B cells. And in vivo experiments determined that PITPNA-AS1 regulated HCC progression in vivo via miR-876-5p/WNT5A pathway. In conclusion, this work shed lights on the modulatory mechanism of PITPNA-AS1/miR-876-5p/WNT5A axis in HCC, which might be pivotal for exploring effective diagnostic biomarkers and treatment strategies for HCC patients.

Background: Recently, emerging studies have identified that MNX1-AS1 highly expressed among variety of cancers and related with worse prognosis of cancer patients. The purpose of this study was to evaluate the relationship between MNX1-AS1 expression with clinical features and prognosis in different cancers. Methods: In this study, we searched the Web of Science, PubMed, CNKI, and Wanfang databases to find relevant studies of MNX1-AS1. Pooled hazard ratios (HRs) and odds ratios (ORs) with 95% confidence intervals (CIs) were applied to explore the prognostic and clinical significance of MNX1-AS1. Results: A total of 9 literatures were included in this study, including 882 cancer patients. The results showed that patients with increased MNX-AS1 expression were more likely to develop lymph node metastasis (OR = 5.616, 95%CI: 3.093 - 10.199, P = 0.000) and advanced TNM stage (OR = 4.625, 95%CI: 2.366-9.040, P = 0.000). Moreover, we demonstrated that patients with high expression of MNX1-AS1 had poor OS in different cancers (HR = 1.976, 95%CI: 1.653 - 2.361, P=0.000). In addition, patients with high expression of MNX1-AS1 suffer from worse prognosis in gastric cancer (HR = 2.385, 95% CI: 1.838 - 3.094, P = 0.000) and lung cancer (HR= 1.959, 95%CI: 1.353 - 2.835, P = 0.000). Conclusions: High MNX1-AS1 expression is significantly associated with unfavorable clinical outcomes and it has the potential to serve as a prognostic biomarker in cancer patients.

Glioma (GM) is a highly lethal human cancer. Circular RNAs (circRNAs) act as an imperative factor in oncogenesis. We aimed to investigate the biological functions and mechanisms of circ‐CDC45 in GM. circ‐CDC45 expression in GM specimens and cell lines was measured by real‐time quantitative reverse transcription polymerase chain reaction. Fisher's exact test and Kaplan‐Meier curves further analyzed its clinical implications. A gain/loss‐of‐function study was conducted to investigate the role of circ‐CDC45 in GM. Additionally, luciferase reporter and rescue assays were performed to unravel the mechanisms of circ‐CDC45. High circ‐CDC45 expression was found in GM specimens and cells, which was tightly related to a larger tumor size, higher world health organization (WHO) stages, and worse survival for patients with GM. Functionally, manipulation of circ‐CDC45 expression strongly affected cell growth, apoptosis, migration and invasion, which suggests the oncogenic function of circ‐CDC45 in GM oncogenesis. Stepwise mechanism studies indicated that circ‐CDC45 sponged and regulated the expression of miR‐516b and miR‐527 to promote cell growth and invasion. Briefly, the regulatory network of circ‐CDC45/miR‐516b/miR‐527 plays a pivotal role in GM tumorigenesis and may act as a potential target for GM treatment.
